Gardenia jasminoides belongs to genus Gardenia of Rubiaceae family.As the medicinal and edible plant resource,it provides medicinal value in its roots,stems,leaves and fruits,which is widely used in medicine,food,cosmetics industry,etc.In this study,seven different G.jasminoides populations in Hunan Province were used as research materials.The genetic diversity of different G.jasminoides populations was studied by 15 phenotypic characters of leaf and fruit with IS SR molecular markers of leaf.HPLC method was used to determine the contents of gardenoside,chlorogenic acid and crocin-I in G.jasminoides from different populations.The results of this study provided a theoretical basis for the evaluation of genetic diversity and comprehensive utilization of G.jasminoides germplasm resources in Hunan Province.The main research results are as follows:1.The results of phenotypic variation analysis showed that the genetic diversity of phenotypic characters of G.jasminoides germplasm resources in Hunan was rich,and there were varying degrees of variation between populations and within populations The variation of phenotypic characters among populations ranged from 8.6%to 39.5%.Among them,the phenotypic differences of crown width,leaf length and fruit fresh weight were greater,and the degree of genetic differentiation was higher.The variation degree of phenotypic character in the populations from high to low is Yueyang population(23.85%),Shuangfeng population(20.87%),Ningxiang population(19.04%),Linwu population(17.82%),Shaodong population(16.99%),Fenghuang population(15.01%),Miluo population(12.48%).2.The result of Principal Component Analysis(PCA)showed that fruit traits contributed most to the genetic diversity of gardenia germplasm resources in Hunan,and the contribution rate was 43.938%;the second principal component was mainly treeshaped traits,and its contribution rate was 10.368%;The third principal component was mainly leaf traits,and its contribution rate was 9.712%.3.Cluster analysis of phenotypic characters showed that the Hunan G.jasminoides populations were divided into three groups.The first group including Ningxiang population、Linwu population、Shaodong population,which have medium fruit weight,similar leaf size and tree shap;The second group including Fenghuang population and Miluo population,with large fresh weight,round fruit shape and long leaves;The third group including Yueyang population and Shuangfeng population,with small fruit shape,light weight and small leaves.4.In this study,seven ISSR primers were used to analyze the genetic diversity of Hunan G.jasminoides populations.64 different clear bands were amplified,60 of which were polymorphic bands,and the percentage of polymorphic loci was 93.75%.The percentage of polymorphic loci among different populations ranged from 25%to 57.81%.Shannon index(I)ranged from 0.15 to 0.32,expected heterozygosity(He)ranged from 0.10 to 0.22,The results showed that G.jasminoides had high genetic diversity.Yueyang population had the most abundant genetic diversity and the strongest ability to adapt to the environment,while Miluo population and Shaodong population had a lower degree of genetic differentiation.The G.jasminoides populations in this study were divided into four groups according to genetic distance clustering,The first was Fenghuang population;The second was Shuangfeng population and Shaodong population;The third group was Ningxiang population,Yueyang population and Miluo population,the last one is Linwu population.The results showed that the closer the genetic distance,the more easily the population clustered into one group.5.The results of HPLC analysis showed that the contents of geniposide,chlorogenic acid and crocin-I in G.jasminoides fruit among different populations were quite different.the content of geniposide varied 53%among different populations,and the Linwu population was the highest(5.98%),and the Shuangfeng population was the lowest(3.90%);The content of chlorogenic acid varied 300%,and the Fenghuang population was the highest(0.12%),and in Miluo population was the lowest(0.03%).The content of crocin-I varied 99.53%,the highest was Fenghuang population(4.23%),and the lowest was 2.12%in Linwu population.By comprehensive evaluation,the fruit quality of G.jasminoides from high to low in seven populations of Hunan province was:Fenghuang population,Miluo population,Ningxiang population,Linwu population,Shaodong population,Shuangfeng population,Yueyang population.
