| Blood biochemical indexes are important indicators of animal’s health and metabolism.To figure out the correlation between blood biochemical indexes and production trait and identify the SNPs,candidate genes and microorganisms which associated with blood biochemical indexes,the phenotypic values of productive traits from 274 Large white pigs(124 boars and 150 sows)at age from 80 to 160-day-old were measured,and blood and fecal samples were collected at age of 80-day-old.The LDH,HDL-C,LDL-C,Glu,TG,SCR,TNF-α,IL-6 and CRP levels in blood were measured,and the genome-wide association analysis of them were carried out.The gene annotation,Go and KEGG enrichment were used to identify the candidate genes.The fecal samples were analyzed by 16 S rRNA sequenceing technology,and the species composition analysis,alpha diversity analysis,beta diversity analysis,species difference analysis,correlation analysis and function prediction methods were applied to obtain the microbiological markers of blood biochemical indexes.The purpose of the present study was to excavate the biological markers which can be used in early selection of Large white pigs.The main results of this study are as follows:1.The correlation analysis results showed that 9 blood biochemical indexes were significantly(P<0.05)or extremely significantly(P<0.01)affected by the sampling month.The levels of blood Glu and HDL-C in boars were significantly lower than sows(P<0.05).The LDL-C were significantly(P<0.05)affected by parity and the level of LDL-C in 1st parity was significantly higher than in 3rd to 5th parity(P<0.05).The highest correlation coefficient was found between TG and Glu(r=0.65)following by HDL-C and Glu,and TG and LDL-C(r=0.51~0.59,P<0.0001).TG was negatively correlated with body length and height(r=-0.16~-0.27,p<0.05),and positively correlated with FCR and corrected 30-100 kg FCR(r=0.14~0.18,p<0.05).LDH was negatively correlated with FCR,corrected 30-100 kg FCR and RFI(r=-0.18~-0.26,p<0.05).sCr was negatively correlated with RFI,ADFI,FCR,corrected 30-100 kg FCR,backfat thickness and corrected 100 kg backfat thickness(r=-0.13~-0.25,p<0.05),and positively correlated with loin muscle area(r=0.13,p<0.05).CRP was negatively correlated with loin muscle area,corrected loin muscle area of 100 kg,loin muscle thickness and corrected loin muscle thickness of 100kg(r=-0.21~-0.23,p<0.01),and positively correlated with FCR(r=0.13,p<0.05).Glu and IL-6 were positively correlated with FCR and corrected 30-100 kg FCR(r=0.12~0.15,p<0.05).IL-6 was positively correlated with RFI and corrected100 kg backfat thickness EBV(r=0.18~0.21,p<0.05).2.A total of 61 SNPs significantly associated with blood biochemical indexes by GWAS analysis were identifed,including 1 SNP in CHR 9 with CRP,2 SNPs in CHR 15 and 4 SNPs in CHR 6 with Glu,5 SNPs in CHR 1with HDL-C,5 SNPs with LDH(1 in CHR 4,1 in CHR 6,1 in CHR 18,and 2in CHR 12),33 SNPs with LDL-C(7 in CHR1,10 in CHR 3,15 in CHR 14,and 1 in CHR 17)and 12 SNPs with TNF-α(1 in CHR 1,1 in CHR 9 and 10 in CHR 8).Two candidate genes related to CRP(CASD1,BET1),2 candidate genes related to HDL-C(JAK2,GNA14),3 candidate genes related to LDL-C(LIPA,APOB,LDAH),3 candidate genes related to LDH(MYH3,TNNT1,TNNI3)and 3 candidate genes related to TNF-α(TIPIN,ZWILCH,ANAPC4)were predicted,respectively.3.The 16 S RNA sequencing results showed that the sampling month,farm,sex and birth weight had significant impacts on the structure of intestinal microbiota.The 80-day-old Large white pigs had two enterotypes(gemmiger and Lactobacillus),which significantly affected the blood LDL-C and TNF α levels(P<0.05).The abundance of Prevotella,faecalibacterium Roseburia and Desulfovibrio were positively correlated,and the Fibrobacter abundance was negatively correlated with blood LDH level.The abundance of Treponema and cf231 were positively correlated,and the subdoligranulum,Eubacterium and Dore abundance were negatively correlated with sCr.These microorganisms can be used as potential intestinal microbial markers related with these biochemical indicators.The functional pathways analysis showed that the differential microbiota related to sCr were significantly enriched in glycan degradation,protein digestion and absorption,amino acid metabolism and other pathways,the differential microbiota related to LDH were mainly enriched in carbohydrate metabolism,nucleotide metabolism,cardiovascular disease and other pathways in boars,the differential microbiota related to Glu were mainly enriched in glycan degradation and apoptosis in sows,the differential microbiota related to HDL-C were mainly enriched in cell movement,lipid metabolism,carbohydrate metabolism and membrane transport in boars,and the differential microbiota related to CRP were mainly enriched in glyceride metabolism.In summary,some blood biochemical indexes asssociated with production traits were identified in the present study.A numbers of SNPs and microbiota markers related with some blood biochemical indexes were also screened by GWAS analysis and 16 S RNA sequencing technology.The results of this study not only provide potential biological markers for the early selection of Large white pigs but also has certain reference value for the clinical research of human related diseases. |
