Anesthesia is an important guarantee in clinical veterinary surgery diagnosis and treatment.Safe anesthesia can alleviate the suffering of the animal,Provide stable physiological indicators,prevent the accidental injury of human and animal,and ensure the progress of teaching and scientific research experiments goes on smoothly.Maintenance anesthesia with isoflurane after intravenous anesthesia is the most commonly used anesthesia method in small animals.Physiological indexes of dogs during induction anesthesia fluctuate widely and the risk rate is high.Therefore,how to carry out scientific and reasonable induction anesthesia,ensure the rapid endotracheal intubation of animals and safely transition to the anesthesia stage is crucial.In this study,alfaxalone,propofol,and zoletil,commonly used in domestic and foreign clinical trials of small animals,were selected as intravenous induction anesthetics before isoflurane maintenance anesthesia,and the advantages and defects of the three anesthetics were compared,so as to provide a scientific and reasonable induction anesthesia scheme for veterinary clinical diagnosis and related scientific research.The experiment consisted of alfaxalone induction group(2.5±0.5 mg/kg),propofol induction group(6±0.5 mg/kg),and zoletil induction group(6±1.0 mg/kg).Each group was applied to 6experimental dogs.Isoflurane with 1.5% concentration was added and maintained for 60 min after successful induction of anesthesia.The effect of induction anesthesia was evaluated by recording the duration of each periods of anesthesia,evaluating the quality of tracheal intubation,inducing anesthesia and recovery.In addition,the safety of the three drugs was comprehensively evaluated by recording the basic physiological indexes such as body temperature,heart rate,respiratory rate,arterial blood pressure,oxygen saturation,end-expiratory carbon dioxide concentration,electrocardiogram waveform and interphase changes,blood routine,biochemical detection and arterial blood gas.Finally,surgical validation and clinical trials were conducted to verify the clinical applicability of the induction protocol.The results showed that the induction time of alfaxalone group was the shortest,and the quality of induction and recovery were the best.The heart rate increased 5 min after intubation,and then decreased gradually.Arterial blood pressure decreased significantly with the increase of anesthesia time,and recovered slowly after 45 min.It has little effect on respiratory system,and there is no significant difference between the indexes and other drugs.The fluctuation amplitude of each index of electrocardiogram was small and all were within the normal range.The induction time of propofol was short,and the quality score was the same as that of alfaxalone.The heart rate increased 5 min after intubation and then gradually decreased,and the decrease amplitude increased 40 min later.5min had a stronger inhibitory effect on respiratory system,and the inhibitory effect on body temperature was stronger than that of the other three groups.Hypotension occurred 30 min after anesthesia,and all electrocardiogram indexes were within the normal range;The induction time of zoletil was the longest,the quality of induction and recovery was medium,the heart rate and arterial blood pressure increased significantly after 0 min of intubation,and then gradually decreased,and the electrocardiogram indexes fluctuated but all were within the normal range.All the three groups were within the normal range of physiologic hematology.In summary,the operation verification and clinical application experiments were conducted with alfaxalone and propofol.In the experiment,the physiological indexes of dogs fluctuated normally and the operation could be completed successfully.Based on the above results,it was concluded that none of the three drugs had significant toxicity to liver,kidney and heart in dogs,and the fluctuation of physiological indexes was transient and recoverable during anesthesia.There was no adverse effect on isoflurane anesthesia in dogs.Alfaxalone and propofol can be used as induction anesthetics alone,alfaxalone has little effect on the cardiovascular system of dogs and are more suitable for intravenous induction anesthesia in dogs with cardiovascular instability.
