Objective Studied in this paper by observing the relationship between the mitochondrial pathways and the related factors Bax,Bcl-2 and Caspase-3 induced by sciatic nerve in rats,and to explore the mechanism of Yunpiheluo Decoction to improve the sciatic nerve injury in diabetic rats.Methods 60 male diabetic rats were randomly divided into model group,Yunpiheluo Decoction low,medium and high concentration group,positive group,and set up normal group to correspond to drug intervention treatment,the whole observation of the general state of rats,Body weight,fasting blood glucose,16 weeks after the detection of nerve conduction velocity,and then dissect the rats,take the side of the sciatic nerve,into the 2.5%glutaraldehyde fixative,the other side of the sciatic nerve into the liquid nitrogen,placed in-80℃ refrigerator.The mRNA expression and protein expression of Bax,Bcl-2 and Caspase-3 in the sciatic nerve of each group were detected by real-time fluorescent quantitative PCR and Western Blot.Result The general condition of diabetic rats such as mental state,activity sensitivity,tail petechiasis,foot ulcer,etc.The rat weight of the treatment group was increased relative to the model group(P<0.05).The FBG of each treatment group was not significantly different from the model group(P>0.05);Rat sciatic nerve structure observed under electron microscope,compared with model group,the damage of high concentration group of TCM to a lesser degree,mitochondria structure clear,nerve fiber degeneration and fracture were significantly decreased,myelin lesions significantly lighter;In the model group,the higher levels of Bax,Caspase-3,and Bcl-2were significantly lower.After the intervention of TCM,Bax,Caspase-3 expression inhibition,Bcl-2expression enhancement.Conclusion1.Using the duplex method(STZ intraperitoneal injection and combined with high fat and sugar feed)mode,the FBG,MNCV tests,indicating DPN in rats model copy success.2.The movement of Yunpiheluo Decoction can improve the extent of axonal deformation caused by the mitochondrial swelling and the separation of the layers of the myelin sheath.3.Yunpiheluo Decoction can significantly reduce the expression of Bax,Caspase-3 mRNA and protein expression of diabetic rat sciatic nerve tissue,and increase the expression of Bcl-2 mRNA and protein.4.Yunpiheluo Decoction on diabetic rats sciatic nerve in the mitochondrial pathway mediated apoptosis and its regulation factor expression regulation function,can effectively alleviate nerve damage,sugar treatment for DPN.
