| Purpose:1 The study collected patients diagnosed with peptic ulcer in the department of spleen and stomach of Liaoning University of Traditional Chinese Medicine,analyzed the clinical information and treatment prescriptions of patients from multiple perspectives,and summarized the incidence characteristics,the correlation of syndrome types and medication rules,as well as the systematic analysis of prescription compatibility rules.2 To verify the role of —— and its functional prescription in different stages,whether the mechanism of action improves the inflammation and pyroptosis of GES-1 cells,promotes ulcer healing and delays the progression of peptic ulcer by inhibiting NLRP3 inflammasome activation.Material and method:1 Clinical study: All the cases were from the spleen and stomach Department of the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine from July2017 to July 2022.50 patients were treated with Western medicine diagnosed as peptic ulcer(including gastric ulcer,duodenal ulcer and complex ulcer).The frequency distribution of the drugs was statistically analyzed using the SPSS25.The frequency of general data was analyzed,including the gender,age,course of disease,solar term of onset and season of onset.Frequency analysis of the characteristics of diseases:including patient inducing factors,classification of ulcer results,combined diagnosis,Chinese medicine syndrome type,tongue coating and pulse image.Distribution analysis of drug use frequency: distribution of drug use frequency and distribution of drug properties.Using IBM SPSS Modeler 18 system to explore the components of core drugs in prescription.For the cluster analysis between drugs,SPSS25 system was used to analyze the statistical drug hierarchical clustering to explore and analyze the combination rules of commonly used drugs in the prescription.The complex network analysis of the drug structure by Cytoscape can form a complex network analysis chart,so as to find and analyze the core prescription composition of the prevention and treatment of peptic ulcer.2.Experimental research: The cell model was established,and CCK-8 was used to select the best drug concentration and serum by drug-containing concentration,which was divided into blank group,model group,deulcer group,qinghong group,antiseptic muscle group,spleen and stomach group,and gastric heqing group.After optimal ethanol concentration,animals were given serum intervention for 24 h.IL-18 and IL-1β inflammatory pro-factor in cell supernatants by ELISA;NLRP 3 and Caspase-1 protein expression by Western-blot;and NLRP 3 and Caspase-1 m RNA in GES-1 cells by RT-PCR.Statistical analysis was performed using SPSS25,one-way ANOVA between groups,P <0.05.Results:1 Through research found that male peptic ulcer patients to more than female,the largest number of winter,red tongue,white vermosy,pulse for the most string.2 Liver and gastric discord is the most common syndrome,in the clinical treatment of liver and gastric disharmony peptic ulcer,the ulcer and liver powder are the most common.3 The optimal molding drug concentration for the cell model was 5% ethanol concentration for 3h.4 IL-18 and IL-1 β protein expression in cell supernatants was detected by ELISA.Compared with the model group,the protein expression of IL-18 and IL-1 in the supernatants of GES-1 cells,Tortefacurid muscle,spleen and stomach,and GES ulcer was significantly lower than that in the model group,which was statistically significant(P <0.05).5 Western-blot method was used to detect the expression levels of NLRP3 and Caspase-1 protein in GES-1 cells after treatment with drug-containing serum.Compared with the model group,the protein expression of NLRP3 and Caspase-1 in the supernatant of GES-1 cells in the heat-clearing and detoxicating group,the putrefaction-promoting granulation group,the spleen-strengthening and stomach-regulating group,and the carbuncle-relieving and dekang group were significantly lower than those in the model group,with statistical significance(P<0.05).6 RT-PCR was used to detect the m RNA levels of NLRP3 and Caspase-1 in GES-1 cells after treatment with medicated serum.Compared with the model group,the m RNA levels of NLRP3 and Caspase-1 in GES-1 cells were significantly decreased in the heat-clearing and detoxicating group,the tissue-supporting and granulation-promoting group,the spleen-invigorating and stomach-invigorating group and the carbuncle-relieving and dekang group,with statistical significance(P<0.05).Conclusion:1 Patients with hepatic and gastric incompatibility are the most common among patients with peptic ulcer disease.According to the analysis of the results of data mining,the core prescription includes 26 drugs,and the core prescription mainly includes prescriptions such as ulcer ulcer,Chaihu and liver dispersion,including the core combination of liver and stomach discord and peptic ulcer.2 By regulating the pyroptosis pathway of NLRP 3 / Caspase-1 cells,the ulcer and the removal can inhibit ethanol to induce pyroptosis of GES-1 cells,so as to improve GES-1 cell damage in human stomach and achieve the purpose of treating peptic ulcer. |
