Objective:To investigate the effects of Bushenshengjing Recipe on the testis,epididymis tissue quality,semen quality,serum sex hormone level and the intracellular signal transduction molecules Smad1 and Smad4 genes of transforming growth factor-? superfamily in oligospermia and asthenospermia of infertility rats,To explore the mechanism of Bushenshengjing Recipe on oligosperrmia and asthenospermia of infertility rats with Kidney-yang Deficiency leaded by adenine,and to provide effective scientific basis for clinical treatment.Methods:120 mature male SD rats were randomly divided into two groups:model group,blank group,high dose group,middle dose group,low dose group and western medicine control group.Each group of 20.The rats in the blank group were given normal saline,and the other groups were given adenine for gavage,and the rats were given once a day for 30 days,Resulting in kidney yang deficiency type weak spermatozoa rat model,each group removed 2 rats,with immunohistochemical detection of testicular and epididymal tissue modeling success.After the success of the model,the high,medium and low dose groups were given different doses of self-made Bushen shengjing prescription,the dosage of the high dose group was 10.22g/ml,the middle dose group corresponding to the concentration of 5.11 g/ml,the low dose group corresponding to the concentration of 2.56g/ml,while the model group and the blank group were given daily lml/100g concentration of saline,the western medicine control group given daily 10ml/kg concentration of carnocatin Injection of each group were fed for 28 days.During the course of the experiment,the rats were observed in the general state(such as color,activity,body,etc.),measured daily drinking water and food intake.At the end of the experiment,the rats were sacrificed,the femoral artery was taken away,the serum was collected,the testis,gonads and accessory glands were harvested and the epididymal semen was extracted.The sperm motility,sperm motility and sperm percentage were measured by computer-assisted sperm quality analysis system.The body weight,testis and epididymis were measured.The expression of Smad 1,Smad 4(LH)and follicle stimulating hormone(FSH)were measured by radioimmunoassay,Smad 1 and Smad 4 were detected by real-time fluorescence quantitative PCR.The levels of Smad 1 and Smad 4 in rat testes were detected by Western Blot and the serum liver and kidney function were measured by enzyme immunoassay.Result:1.Compared with the model group,the difference between the sperm count and the sperm motility test group was statistically significant(P<0.05),compared with the model group,the high dose group,the middle dose group,the low dose group,the western medicine(P<0.05).The sperm count and sperm motility in the high dose group were significantly higher than those in the middle dose group,the low dose group and the western medicine control group(P<0.05),and the sperm motility and sperm motility were significantly higher in the control group(P<0.05),but there was no significant difference between the middle dose group and the western medicine control group(P>0.05),the difference was statistically significant(P<0.05)2.The effect of Bushen Shengjing Decoction on the gonad and gonadal gland of neonatal deficient rats was significantly lower than that of the blank group(P<0.05),and the difference was significant(P<0.05).There was no significant difference between the low dose group and the model group(P>0.05),but there was no significant difference between the high dose group and the middle dose group and the western medicine control group(P>0.05)There was no significant difference between the dose group and the western medicine control group(P>0.05).Compared with the blank group,the wet weight of the testes and the wet weight of the epididymis were significantly lower than those in the control group(P<0.05).Compared with the model group,the high dose group,middle dose group and western medicine control group were significantly different(P<0.05).There was no significant difference between the low dose group and the model group(P>0.05).Compared with the western medicine control group,there was no significant difference between the low dose group and the control group No significant difference(P>0.05).3.Smadl and Smad4 were detected by immunohistochemistry.Smad 1 and Smad4 were expressed in the spermatogenic cells of all the testes in the blank group and the model group.The Smadl immunoreactive substance was located in the cytoplasm.Negative,stromal cells and supporting cells were not positive expression;Smad4 immunoreactive substances in the nucleus,the cytoplasm was negative,support cells,stromal cells and other levels of spermatogenic cells were not positive expression.Compared with the model group,the expression of Samdl in the other groups was significantly lower than that in the control group(P<0.05).The expression of Samdl in the high dose group was significantly lower than that in the middle dose group,low dose group and western medicine control group(P<0.05).There was no significant difference between the middle dose group and the western medicine group(P>0,05).Compared with the model group,the expression of Samd4 in the other groups was significantly higher than that in the control group(P<0.05).The expression of Samd4 in the high dose group was significantly higher than that in the middle dose group,low dose group and western medicine control group(P<0.05),while the middle dose group and the western medicine control group The difference was not statistically significant(P>0.05)4.Serum levels of testosterone(T)were decreased and LH and FSH levels were increased in the model group.The levels of serum testosterone(T)in the high,middle and low dose groups were significantly higher than those in the control group(P<0.05).Compared with the model group,the levels of serum testosterone(T),LH and FSH in the other groups were significantly higher than those in the control group(P<0.05),and the difference was significant Significant differences were statistically significant(P<0.05).5.The expression of Smadl and Smad4 mRNA in the testes of each group showed that Smadl mRNA expression was observed in the testes of each group,and Smadl mRNA expression was inhibited in the model group.MRNA expression was up-regulated,and the expression of Smadl mRNA was up-regulated in the high dose group of Kidney-spermatogenic rats,and the expression of Smad4 mRNA was inhibited.6.The liver and kidney function was detected by enzyme immunoassay.The appearance,histopathological examination and ultrastructural observation of liver and kidney were observed in all groups,and there was no significant change in serum test.The treatment of Kidney essence is less and less severe infertility rats safe and reliable,no obvious adverse reactions.Conclusion:1.Self-made Bushen Shengjing side can effectively increase the semen density and effectively improve the sperm viability and vitality,and by regulating the secretion of testosterone,increase the quality of testes and append glands,so that the spermatogenic environment improved,less weak sperm The treatment has a better effect.2.Bushenshengjing can promote the growth and metabolism of rats and the development of testes and epididymis in rats.3.Bushenshengjing can promote luteinizing hormone(LH)release,improve testosterone(T)concentration,reduce estradiol(E2)and reduce the concentration of estradiol(E2)Content,regulation of hypothalamic-pituitary-testicular axis function,improve the gonadal axis dysfunction.4.The expression of Smad1 in the testes of Kidney-deficient and under-weak rats was significantly increased,and the expression of Smad4 in the testes of Kidney-deficient and less-weak rats was significantly increased and the mRNA of Smad 1 was up-regulated Expression,inhibition of Smad4 mRN A expression,indirectly promote the generation of sperm to achieve the purpose of treatment of male infertility.5.There was no significant change in liver and kidney function in the serum of each group by enzyme immunoassay.It was proved that self-made Kidney-sosperm was safe and reliable without any adverse reaction.
