| Objective To study the effects of Buyanghuanwu Decoction on the brain’s Notch signaling pathway of the rats with depression after cerebral ischemiaMethods To make a focal cerebral ischemia modle in rats with suture method firstly,then dividing the rats into5groups randomly, namely control group,MCAO group, PSD group,PSD+Buyanghuanwu decoction group and PSD+fluoxetine group. To add the CUMS(Chronic Unpredictable Mild Stress) combining with solitary to these rats in PSD group,PSD+Buyanghuanwu decoction group and PSD+fluoxetine group to compound the PSD model.From the first day of adding CUMS combining with solitary to the rats,each group was intragastric administration one time every day.To weigh all the rats and do open-field test for all the rats to estimate wheather the depressive symptoms of the rats were improved. And to do immunohestochemistry with the rats’brain for the4indexes which are hes1ã€hes5ã€jag1and notchl in Notch signaling pathway.To observe the effects of Buyanghuanwu Decoction on the Notch signaling pathway. Results1ã€Open-field Test result:Comparing with PSD group,the trajectory distance of the Open-field Test of PSD+Buyanghuanwu decoction groupã€PSD+fluoxetine groupã€MCAO group and the control group are not different in the two time points of the7-th day after operation and before the experiment,but on the21-st day,the trajectory distance of these four groups add obviously, but between PSD+Buyanghuanwu decoction group and PSD+fluoxetine group,the difference is not obvious.2ã€Weight change result:Weight difference (weight difference between the rats’ weight on the21-st day after operation and on the day before the trial)showed:Compared with the PSD group, the other four groups’ weight are much higher than the former, the differences are obvious, but between the PSD+Buyanghuanwu decoction group and PSD+fluoxetine group,the difference is not obvious. Compared with the PSD group,the weight ratio (the rats’ weight difference/the rats’ weight before the experiment) of the other four groups increased significantly,there is a sharp difference.3ã€Notch signaling pathway immunohistochemical statistical analysis results showed:In the same time point, compared with the PSD group, the average optical density of notch1, jag1, hes1and hes5of Notch signal pathway in the brain of the PSD+Buyanghuanwu decoction group and PSD+fluoxetine group increased. The effects of Buyanghuanwu decoction and fluoxetine are similarly the same. With the progress of the time, compared with the PSD group, the MCAO group’s average optical density of the four indicators in Notch signaling pathways also increased.Conclusion1ã€Buyanghuanwu decoction which is similar with fluoxetine can improve depressive symptoms of the rats with depression after cerebral ischemia.2ã€Buyanghuanwu decoction can enhance expression of notchl, jagl, hesl and hes5in Notch signaling pathway in the brain of the rats with depression after cerebral ischemia to improve the depressive symptoms of the rats with depression after cerebral ischemia. |
