Objective(s):The purpose of this project is to observe the effects of maternal inflammatory stimuli during pregnancy on the kidney development of the offspring,and from maternal disease "programmed" point of view to clarify maternal inflammatory stress has an adverse effect on fetal kidney development.Methods:SD male and female rats were used to establish a pregnant rat model by 1:2 mating.The next day after mating do female vaginal examinations,vaginal suppository show modeling success.Forty-eight pregnant rats as experimental subjects were randomly selected and were equally divided into three groups by random number tablecontrol group,LPS group and blank group,sixteen rats in each group.The control group was given intraperitoneal injection of sterile saline.LPS group was given intraperitoneal injection of LPS.The blank group was given no treatment.Three groups of pregnant rats were subdivided into 2 groups again and were sacrificed on the 13th and 20th day respectively.The CRP levels of pregnant rats in each group were measured,the serum creatinine levels and the serum urea nitrogen levels of pregnant and fetal rats in each group were measured,and the data of each group were compared and statistically analyzed.Light microscopic observation of renal and placenta pathological changes after HE staining was completed.During the experiment,rats were fed normally at(24 ± 1)?,feeding and drinking water freely.Closely observe pregnancy,abortion or not,mental status,activity frequency,appetite and survival rates.Results:1.In this study,a total of 48 pregnant rats were randomly divided into three groups:control group,LPS group and blank group,16 in each group.The control group was given intraperitoneal injection of sterile saline.The LPS group was given intraperitoneal injection of LPS.And the blank group has no injection.There was no significant difference in fetal growth index(fetal quality and placental quality)between the blank group and the control group on the 13th and 20th day of pregnancy(P>0.05).Compared with the blank group and the control group,the LPS group had lower fetal and placental quality,and the difference was statistically significant(P<0.05).2.In the 13-day and 20-day group,measuring and comparing their CRP levels,there was no significant statistical difference in CRP levels between the control group and the blank group(P>0.05).Compared with control group and blank group,levels of CRP in LPS group were significantly increased,and the difference was statistically significant(P<0.05).3.In the 13-day group,measuring and comparing their renal function parameters,serum creatinine and urea nitrogen levels,there was no significant statistical difference in biochemical indexes of renal function between the control group and the blank group(P>0.05).Compared with control group and blank group,levels of Scr and BUN in LPS group were significantly increased,and the difference was statistically significant(P<0.05).4.In the 20-day group,measuring and comparing their renal function parameters,serum creatinine and urea nitrogen levels,there was no significant statistical difference in biochemical indexes of renal function between the control group and the blank group(P>0.05).Compared with control group and blank group,levels of Scr and BUN in LPS group were significantly increased,and the difference was statistically significant(P<0.05).Fetus were taken out,measuring the indexes of fetal kidney function,the serum creatinine and urea nitrogen levels.there was no significant statistical difference in biochemical indexes of renal function between the control group and the blank group(P>0.05).Compared with control group and blank group,levels of Scr and BUN in LPS group were significantly increased,and the difference was statistically significant(P<0.05).5.Light microscope observation after HE staining:in the blank group and control group,the kidney structure of pregnant rats and fetal rats was clear,the glomerulus was normal,there was no invasion of inflammatory cells and no obvious lesion.In the LPS pregnant group,the glomerulus capillaries of were congested and swollen,the glomerulus stuctre was disordered,and renal tubular epithelial cells are swollen and degenerated,with narrow lumens,interstitial inflammatory cell infiltration,and some even with segmental sclerosis.In the LPS fetal group,the glomerulus structure was disordered and there was a large number of red blood cells and inflammatory cells infiltrated in the interstitium.In the blank group and control group,the placental tissue had clear structure and gradation,and the cell morphology was regular.In the 13-day LPS group,the organizational structure was disordered,the level boundaries were unclear.Small blood vessels that were hardened and hyperemia were observed.The structure of the 20-day LPS group was more disordered than that of the 13-day group,and the levels could not be clearly distinguished,and there was even a large number of inflammatory cell infiltration.Conclusion(s):1.Compared with control group and blank group.levels of CRP in LPS pregnant group were significantly increased,and the difference was statistically significant(P<0.05).The successful modeling suggests that LPS can lead to maternal inflammatory responses,and changes in renal function may be related to inflammatory stress.2.The placental quality and fetal quality in the LPS group were significantly lower than those in the blank group and control group(P<0.05).The placental structure in the LPS group was more disordered than that in the other two groups,suggesting that maternal long-term exposure to inflammatory stimuli could lead to placenta structural damage,inhibit fetal rat development and affect the development of the embryo.3.Compared with control group and blank group,levels of Scr and BUN in LPS pregnant and fetal group were significantly increased,and the difference was statistically significant(P<0.05).Meanwhile,the pathological changes of kidney structure in LPS group were more obvious than those in control group and blank group.It indicated that inflammatory stress can cause kidney dysplasia and structure change in pregnant rats and their offspring,providing the basis for maternal inflammatory stimuli causing offspring kidney development dysplasia.It can provide reference for the prevention and treatment of early kidney disease.Also it has a certain clinical application value.
