SECTION 1 :The Content , States of Testosterone in Prostate andRelations with BPHObjectives : Androgen exists in prostatic tissue , but its approchs that effect on prostate in detail have been uncertain . Androgen have two physical states in human body : free and reversibly bound to proteins . In the investigation , we assay the content and distribution of the total , bound and free androgen in prostate of 16 young control and 25 cases of BPH , analyse their physiological effects on prostate and relations with BPH.Methods : The 16 normal young prostatic samples were obtained from young donors died in accute accidents . The benign prostatic hyperplasia tissue removed by superpubic prostatectomy in 25 patients with BPH . Cytosol and nucleusof prostatic tissue were fractionated two parts : in one , total testosterone were measured with 125I-labelled radioimmunoassay , in the other , free and bound testosterone were separated by ether stripping in 2 minutes . To centifugate and remove ether , and bound T were measured by radioimmunoassay .Results: There are two states of testosteone in prostate, the bound is more than the free . In cytosol , total , bound , free testosterone of BPH group were 3.947?.880, 2.180?1.307, 1.767 ?1.660 (ng/g tissue wet weight) , those of young control group were 2.940 ?0.656 , 2.447 ?0.318 > 0.493 ?.415 (ng/g tissue wet weight) . Comparison between the two groups was not significant (P) 0.05). The ratio of free to total in BPH group , 0.396 ?.182 , was significantly higher than that in young control group , 0.152?0.130 (P (0.05) . In nucleus , total , bound , free testosterone of BPH group were 2.571+ 2.268, 1.374 ?.254, 1.197 ?.088 (ng/g tissue wet weight) , those of young control group were 1.663 ?.628, 1.137 ?.072,0.526 ?.481 (ng/g tissue wet weight). BPH group were a little higher than young control group , but comparison between the two groups was not significant (P) 0.05).Conclusions : Two physical states of testosterone exist in human prostatic tissue : free and reversibly bound . The existence of bound testosterone favor the cumulation of testosterone in prostate , free testosterone can be transfered to dihydrotestosterone or bound with androgen receptor to initiate hormone action . To compare BPH group and young control group , total and bound testosterone maintained high level and free testosterone had trend to rise . The two states of testosterone have importantphysiological effect on prostate and relations with BPH.
