White lesions of vulva is a common disease which is difficult to treat,there is no satisfactory method to treat it at present. Ultrasound treatment is a therapy developing quickly.We study effect of ultrasound on vulva skin to investigate the possibility and mechanism in treating white lesions of vulva by ultrasound.1.Effect on vulva skins of rabbits by ultrasound in different doses We divide 25 rabbits into 5 equal groups and irradiate them with CZF-1 ultrasound therapy model machine,irradiating vulva dermis of 5 groups by 1,2,3,4 and 5W respectively and observing pathologic changes of irradiation areas.No evident change in 1 and 2 group.But hyperemia and edema are found on vulva skins 1 hour later in 3,4 and 5 group.Hyperemia and edema are evident as irradiation dose increases.Skin temperature is higher than normality.Irradiating with 3W,hyperemia and edema disappear gradually on first day,as irradiating with 4 and 5W, hyperemia and edema begin to disappear on second day.No hyperemia and edema found on fourth day.Skin temperature is also normal.No coagulative necrosis in all groups.No distinctmicroscopic change in 1 and 2W irradiation group,but edema and gentle hyperemia present in papillary layer of dermis of 3,4 and 5W irradiation groups 1 hour later.Hyperemia and edema disappear 4 days later, chronic inflammatory cell may exist.These results suggest that therapy effect on dermis can be preduced by ultrasound irradiation in certain dose.No coagulative necrosis and epidermis trauma.We presume that it is effective and safe to use ultrasound to treat skin and mucous membrane diseases of vulva.2.Pathologic changes of vulva skin of rabbits after ultrasound irradiationWe divide 50 rabbits into 5 equal groups(4 irradiation groups and 1 control group),irradiating vulva dermis in irradiation groups with CZF-1 ultrasound therapy model machine,irradiation power 4W.We take samples on 1,4,7 and 14 days after irradiation. There are hyperemia and edema at irradiation area 1 day later, epidermis is complete and a little rigid,skin temperature is higher than normal skin.After 4 days,there are no hyperemia and edema,a little rigid epidermis and normal skin temperature.Vulva skin show normal 7 days later.Microscopic changes:edema and gentle hyperemia exist in papillary layer of dermis 1 day later.Hyperemia and edema disappear 4 days later,chronic inflammatory cell may exist.Hypertrophy of capillary vessel appears 7 days later.The skin resume totally 14 days later.Electron microscopic changes:1,4 and 7 days after ultrasound irradiation,endothelialcell of blood vessel exist mitochondria swelling and more micropinocytotic vesicle,fibroblast have mitochondria swelling and rough endoplasmic reticulum dilatation,nerve fibre also exist mitochondria swelling.These results suggest ultrasound is in favor of hypertrophy and rehabilitation of microvasculature,collagen fibrils and nerve ending.We presume that it is possible to use ultrasound to treat white lesions of vulva.3.Changes of ecNOS protein and mRNA in vulva skin of rabbit after ultrasound irradiationWe use immunohistochemistry and hybridization in situ to investigate changes of ecNOS protein and mRNA in vulva skin of rabbit after ultrasound irradiation. After irradiation, ecNOS protein and mRNA are high distinctly on 4 day,resume normal on 7 day.These results suggest that ultrasound irradiation increase NO by its biological effect.NO relax microvessel,enhancing perfusion of capillary vessel,improving function of microvessel and nutrition of local tissue.These are not only benefit to elimination of denaturation and necrosis but also are good for regeneration and restoration of pathologic tissue.We presume that it is possible to use ultrasound to treat white lesions of vulva by improving function of microvessel and nutrition of local tissue.as well as NO is in favor of regeneration of nerve ending.4.Changes of VEGF protein and mRNA in vulva skin of rabbit afterultrasound irradiationWe use immunohistochemistry and hybridiz...
