Objective To study the relationship between selenium and Helicobacter pylori(HP). Methods Influence of selenium on Hp was studied in vitro by morphology, index of biochemistry and influence on vacuole toxin of HP was studied by agarose gel electrophoresis, Sequence analysis, Western blotting and cell culture. Effects of selenium on gastric mucosal lesions of BALB/C mice was studied by HP infection and immunological method. Expression of PCNA, CD44V6, P16 and C-met were investigated in 126 cases gastric mucosallesions by using immunohistochemistry. Hp was examined by both of W-S method and RUT,selenium level in gastric mucosal was determined by spectrofluorometer. Rusults Our research suggested that Effected by given concentration of selenium, the number of HP colony was decreasing with increasing of selenium concentration, the color of colony vary from white to brown. we also discovered that selenium can decrease hemotolysis and effects function of vacuole toxin of HP . Morphological conversion from spiralto coccoid forms occurred when HP was cultured with selenium and most of coccoid forms had fragmentary flagella and pilus; the effection of selenium to activity of urease, catalase, alkaline phosphatase is not exist or not evidence; selenium enable pET32a-vacA-E.coli BL21 which can express VacA produce yellow lipochrome, and the color become deeper with increasing of concentration of selenium; selenium can decrease expression of recombined VacA by restrain the growth of pET32a-vacA-E.coli,but can not influence activity and gene sequence of VacA; selenium can enhance the suppression effect of recombined VacA for gastric cancer cell, depress the index of cell multiplication and the forming rate of colony, decrease the number of cell in S period, increase total of cell in G period; After purified, the recombined VacA can be identified by VacA antibody. Mice test confirmed that:a given concentration of HP can induced gastric mucosal lesions and precancerosis, down regulating CD4/CD8 in blood of mice; selenium can effectively prevent gastric mucosal damage which induced by HP and enhance the activity of CD4 cell, up regulating CD4/CD8. HP positive groups was compared with HP negative groups in gastric mucosal lesions expression of P16 in HP positive groups was lower than that in HP negative (P <0.01), expression of CD44V6,PCNA and C-met in HP positive groups were all higher than those in negative groups (P<0.01). Expression of P16 was negative correlated with expression of CD44V6 , PCNA and C-met in HP-infected gastric mucosal lesions (rs=-0.6971, P<0.01; rs=-0.4811, P<0.01; rs=-0.6075, P<0.01); Expression of CD44V6 were positive correlated with expression of C-met and PCNA (rs=0.6550, P<0.01; rs=0.4977, P<0.01). Selenium level in HP-infected gastric mucosal was Significantly higher than normal and the level will fall with deteriorism of gastric mucosal lesions. Selenium level in HP-infected gastric mucosal lesions Was negative correlated significantly with Expression rat of CD44V6, C-met and PCNA ( P<0.05 ), but was positive correlated with expression rat of P16 (P<0.05). Conclusions Selenium can influence morphological conversion, and decrease hematolysis and effects the function of vacuole toxin of HP. Selenium enable pET32a-vacA-E.coliBL21 which can express VacA produce yellow lipochrome, and the color become deeper with increasing of concentration of selenium; selenium can decrease expression of recombined VacA by restrain the growth of pET32a-vacA-E.coli,but can not influence activity and gene equence of VacA; selenium can enhance the suppression effect of recombined VacA for gastric cancer cell; Selenium can effective pervert HP-infected gastric mucosal damaged by boost CD4 cell activity, enhance immunity of body and depress the down regulating of CD4/CD8; HP infection can enhance expression of CD44V6, PCNA , C-met in gastric mucosal lesions and depress expression of P16; selenium can direct or indirectly depress the expression of CD44V6, PCNA , C...
