Objective To study the effect of Yangxin granule on serum C-reactive protein,P-selectin and the expression of arteria carotis communis P-selectin mRNA, in vulnerable plaque rabbit model with p53 gene transfection,to investigate its mechanism of action at level of molecular biology, to provide a reliable theoretic basis for this therapy.Methods Carotid artery wall injuries were introduced an intravascular balloon in rabbits before they were fed on high fat diet for 8 weeks.At the end of the 8th week,recombinant adenovirus vector by transfecting p53 gene were injected to carotid segments,rabbits were randomly divided into four groups:Model control group,Yangxin granule group, simvastatin group and blank group (6 in each group).Administration for two weeks and plaque rupture because of pharmacological triggering.At the end of the 8th and 10th weekend,the content of serum CRPand P-selectin was examined, observer the expressing change of arteria carotis communis P-selectin mRNA by RT-PCR.Results 1.Compared with blank group,the content of serum CRP in other groups obviously increased at the end of the 8th week(P<0.01).Compared with Model control group,the content of serum CRP declined in treatment groups at the end of 10th week(P<0.01).There was not significant difference between Yangxin granule group and simvastatin group (P>0.05).2. Compared with blank group,the content of serum P-selectin in other groups obviously increased at the end of the 8th week(P<0.01).Compared with Model control group,the content of serum P-selectin declined in treatment groups at the end of 10th week (P<0.05).There was not significant difference between Yangxin granule group and simvastatin group (P>0.05).3.To compare with blank group,the model control group can increase regulation expression of model P-selectin mRNA (P<0.01),to compare with the model control group,the treatment group can down regulation expression of model P-selectin mRNA(P<0.01), There was not significant difference between Yangxin granule group and simvastatin group (P>0.05). Conclusion Yangxin granule remarkably decreased the content of serum CRP and P-selectin,down regulation expression of model P-selectin mRNA in vulnerable plaque rabbit model with p53 gene transfection,which might be one of the mechanisms for stabilizing vulnerable plaque.
