Effect Of Xinwei Granule On Stat3,Survivin,Egfr,Ptenin Rats With Precancerous Lesions Of Gastric Carcinoma

Objective:To investigate the effect of Xinwei granules on the STAT3,Survivin,EGFR,PTEN in rats with precancerous lesions of gastric carcinoma and to explore the mechanism of Xinwei granules and Yiqi Yangyin Huoxue Jiedu principle in the treatment of PLGC.Method:120Wistar rats were randomly divided into model group, normal control group, Xinwei granules groups (including low dose group, medium dose group and high dose group) and Weimeisu group of20. The rat model of precancerous lesions of gastric carcinoma(PLGC)was established by other molding methods in which MNNG was mainly used,16weeks. After the rat model was establi shed,treating12weeks, Weimeisu was given1.0g/kg per day in Weimeisu group; According to the adult clinical dose(20g·d) for the conversion, low,medium and high dose groups of Xinwei granules were respectively given1.254g/kg·d,2.508g/kg·d,5.016g/kg·d per day.Model group was given the equal volume normal saline. After16weeks, to observe the pathological changes with the light microscope in rat gastric mucosa as well as PTEN,EGFR levels by ELISA method and STAT3mRNA,Survivin mRNA expression by Realtime-PCR method.Results:(1)The STAT3expression level of model group is significantly increased compared with normal control group,Xinwei granules groups effectively restrain the level of STAT3,the influence of medium dose group is optimal, better than other groups (P<0.05).The efficacy of high and low dose group of Xinwei granules and Weimeisu group is similar to each other, there is no obvious difference among the three groups (P>0.05)(2)The Survivin expression level of model group is significantly increased compared with normal control group, Xinwei granules groups effectively restrain the level of Survivin,the influence of medium dose group is optimal, better than other groups (P<0.05).The efficacy of high and low dose group of Xinwei granules is better than Weimeisu group, there is obvious difference among the three groups (P<0.01)(3)The EGFR level of model group is significantly increased compared with normal control group,Xinwei granules groups effectively restrain the level of EGFR. The efficacy of high medium,low dose group of Xinwei granules is better than Weimeisu group, there is obvious difference among the three groups(P<0.01).The efficacy of low and medium dose group of Xinwei granules is similar to each other, there is no obvious difference among the three groups (P=1)(4)The PTEN level of model group is significantly reduced compared with normal control group,Xinwei granules groups effectively increase the level of PTEN,the influence of medium dose group is optimal, better than other groups (P<0.05),The efficacy of high and low dose group of Xinwei granules is better than Weimeisu group, there is obvious difference among the three groups (P<0.01)Conclusion:Xinwei granules can improve the pathological conditions of gastric mucosa in rat with PLGC and prevent the progress of PLGC by reducing STAT3mRNA, SurvivinmRNA expression and EGFR level, increasing the expression level of PTEN,Xinwei granules and Yiqi Yangyin Huoxue Jiedu principle can intervene PLGC.
