Objective:To study the effect of Xinwei granule and its compositions of prescription on the Wnt signal transduction pathway in rats with precancerous lesions of gastric carcinoma.Method:140 Wistar rats were randomly divided into 3 control groups including normal control group,model group and Wei Fuchun group.4 treating groups includeing benefiting Qi and nourishing Yin group,benefiting Qi and nourishing Yin and promoting stasis group,benefiting Qi and nourishing Yin and detoxifying group and Xinwei granule group.Totally 7 groups.To establish The rat model of precancerous lesions of gastric carcinoma(PLGC),many pathogenic factors was mainly used together with MNNG.The corresponding drug was given to every group.But Physiological saline were given in normal control group and model group.After 16 weeks,to observe the pathological changes in rat gastric mucosa as well as Wnt-1??-catenin,APC,Axin expression.Result:1.The expression of Wnt-1,p-catenin in model group,Weifuchun group and Chinese medicine group significantly increased,and the expression of APC,Axin significantly decreased compared with normal control group(p<0.05);2.The expression of Wnt-1,?-catenin decreased and expression of Axin increased in Traditional Chinese medicine groups and Weifuchun group compared with the model group,and the differences between groups were statistically significant(p<0.05);3.The expression of APC,Axin in model group apparently decreased when compared with Weifuchun group(p<0.05);4.The expression of Wnt-1 decreased and expression of Axin and ?-catenin increased as comparing Xinwei granule group,benefiting Qi and nourishing Yin and promoting stasis group,benefiting Qi and nourishing Yin and detoxifying group with benefiting Qi and nourishing Yin group.(p<0.05);5.Xinwei granule group had lower expression of Wnt-1 and higher expression of APC and Axin as compared with benefiting Qi and nourishing Yin and promoting stasis group,and the difference was of statistically significance(p<0.05);6.Xinwei granule group had lower expression of Wnt-1 and higher expression of APC and Axin as compared with benefiting Qi and nourishing Yin and detoxifying group,and the difference was of statistically significance(p<0.05);7.No significant difference of the expression of Wnt-1,?-atenin,Axin and APC appeared between benefiting Qi and nourishing Yin and promoting stasis group and benefiting Qi and nourishing Yin and detoxifying group.Conclusion:1.The expression of Wnt-1,?-atenin in model rats of PLCG was increased and the expression of Axin,APC w,as decreased significantly.2.The expression of Wnt-1,?-atenin in Traditional Chinese medicine groups were reduced and the expression of APC,Axin was increased compared with the model group,and gastric mucosal pathology was significantly improved;3.Benefiting Qi,nourishing Yin,promoting stasis and detoxifying group was of the most significant therapeutic effect in all the compositions of prescription group of Xinwei granule,which made better effect on Wnt-1,?-catenin,Axin than that of the control drug Weifuchun;4.All the compositions of prescription group of Xinwei granule made better effects on APC than Weifuchun group;5.It shows in this study that the theareuputic effect of benefiting Qi,nourishing Yin,promoting stasis and detoxifying group(Xinwei granule group)is better than that of supplementing Qi and nourishing yin and removing blood stasis group,supplementing Qi and nourishing yin and detoxifying group,and the supplementing qi and nourishing Yin group.It has a synergistic effect between“law”and“law”,but different combinations of“law”also have some differences in the effect of Wnt pathway.
