Anti-inflammatory Effect And Mechanism Of Volatile Oil Of Basil Leaves

Perillae leaf is one kind of commonly used Chinese herb which belongs to the category of treating Exterior syndrome. It has the function of dispelling exterior and cold evil,and relieving Qi stagnation in Stomach. It is frequently used for treating Exterior Syndrome and some diseases associated with inflammation. The volatile oil is one of the major effective components of this herb,but there is few pharmacological researches about its anti-inflammatory function,let alone its mechanism. According to the intrinsic relations between Exterior Syndrome and inflammation,acrid taste and volatile oil,this dissertation firstly studied the anti-inflammatory effect and mechanism of volatile oil of Perillae leaf (VOPF),from the level of entire animal,tissue and organ,cell and molecule thoroughly and systemically,by means of advanced modern technology such as cell culture,ELISA,flow cytometry,radioimmunoassay,HPLC etc.The results showed:l.VOPF exhibited definite anti-inflammatory effects on many animal models,whether the acute model characterized by increased capillary permeability,swelling,emigration of WBC or the chronic model characterized by hyperplasia of granulation. Especially for the acute inflammatory model,the effect is striking. It proved that the anti-inflammation effect of VOPF is significant and clear-cut. VOPF is one of the primary substantial basis of its relieving exterior syndrome function.2. The anti-inflammatory mechanism of VOPF is as follows:Inhibition the production and activity of COX2 in themetabolism process of AA and decreasing the production of PGE2 by this way;Meanwhile reducing the content of LTC which is produced by AA through the Lipoxygenase pathway. Inhibition the free-radical production such as NO,MDA. Decreasing the content of proinflammatory factors as TNFa .For chemotactic factor IL-8,VOPF appeared inhibited tendency. Preventing the expression of ICAM-1 in EC,then controlling the adhesion between EC and WBC. Inhibition the activation of NF- K B which is the central link of the inflammation.3. The realization of anti-inflammation of VOPF is not by enhancing the function of HPA,but is some kind of direct influence.4. The results indicated that maybe VOPF could affect the Toll signal trarisduction pathway,then inhibit the activation of NF- K B,the center of inflammation. And further prevent the gene expression and protein secretion of adhesive molecule,cytokine,some enzymes associated with inflammation such as PLA2,COX2,iNOS etc.At last decrease the synthesis and release of inflammatory mediator PCs,Us,NO.
