Cortex Magnoliae Officinalis is one of the Traditional Chinese Medicine incommon use. It is named as Houpo. Houpo which produced in Hubei Enshi is highquality. Enshi Houpo is called "Zhi You Houpo", which inside surface color is deeppurple-brown, the color of its cut is brown, the cortex is thick, the aroma is strong, thetexture is oily. Its source plant is Magnolia officinalis Rehd. et Wils. In order to studythe inherent quality of Enshi Houpo, we investigated the efficacy and its materialfoundation, analyzed the content of the active substances, compared the content ofmagnolol and honokiol in Cortex Magnoliae Officinalis produced in different habits,and established the HPLC fingerprint and ISSR fingerprint of Enshi Houpo.We investigated the discussions about Houpo's efficacy in the books on Chineseherbal medicine and the achievements of component analysis and pharmacologicalresearch on Cortex Magnoliae Officinalis. The efficacy of Cortex MagnoliaeOfficinalis is "descending qi, relieving flatulence, dissipating dampness andunblocking full". The active substances can be classified as phenols, volatile oil andalkaloids. Again, the alkaloids are poisonous components of Houpo.Determining the content of magnolol and honokiol in Cortex MagnoliaeOfficinalis by HPLC with methanol-water (78:22) and the detective wavelength was294 nm. For Enshi Houpo, the tree is elder, the content of magnolol and honokiol islower. The content of the compounds in the root cortex is highest while the stemcortex upside is lowest. Fahan could increase the content of magnolol and honokiolgreatly. The content of magnolol and honokiol in Enshi Houpo is highest comparingwith Houpo produced in other habits.Determining the content of volatile oil of Enshi Houpo by the determination ofvolatile oil that prescribed by Ch P and by GC-MS. The content of volatile oil of thetrees which is 5 years old and 20 years old is higher, while the trees of 10 and 15years old are lower. The content of compounds in the branch cortex is highest, whilethe stem cortex downside is lowest. The content of oxide derivatives ofmonoterpenoids and sesquiterpenoids is highest in the stem cortex of M. officinalis of 15 ages.Determining the content of alkaloids in Enshi Houpo by the non-water titration.The tree is elder, the content of alkaloids is lower. The content of alkaloids in the rootcortex is highest while the stem cortex is lowest. Fahan could decrease the content ofalkaloids.Determining the lixiviating content by the determination prescribed by Ch P.The lixiviating content in the stem cortex and the branch cortex of Magnoliaofficinalis is higher while the root cortex is lower. The tree is elder, the lixiviatingcontent is downtrend. Fahan could increase the lixiviating content greatly.HPLC method was used with methanol-water(67:33) as the mobile phase atroom temperature. The chromatographic column was Cosmosil C18-MS-Ⅱ(4.6mm×250 mm,5μm ). The flow rate was 1 mL.min-1, and the detection wavelengthwas 230 nm. The chromatograms of 10 individuals ofEnshi Houpo(the stem cortex ofM. officinalis) were recorded and compared. The comparability of the chromatogramsis high(>0.92). The HPLC fingerprint for reference of Enshi Houpo(the stem cortexof M. officinalis) is made. There are five characteristic absorption bands. The arearatio of peak 5 to 4(characteristicⅠ) and the area ratio of peak 4 and 5 to the totalpeak areas(characteristicⅡ) are characteristics of the HPLC chromatogram of EnshiHoupo(the stem cortex of M. officinalis).PCR amplification of Cortex Magnoliae Officinalis genomic DNA was based onInter-simple sequence repeats (ISSR). Two ISSR primes can be used to construct theISSR fingerprint of M. officinalis produced in Hubei Enshi. There are twocharacteristic bands in the amplification product with ISSR prime 826, whichmolecular weights are 950 bp and 550 bp. The ISSR marker is an effective methodfor the cultivar and habit identification of Cortex Magnoliae Officinalis.
