The purpose of this study was to screening the genetic markers can be used for selecting elit long distance runners and to prediction for effect of aerobic endurance training and HiHiLo training in HEPC、FPN1 and other genes by adopting PCR-RFLP, MALDI-TOF-MS and Gene scan technology.Part I:The feasibility of HEPC、FPN1 and other genes as genetic markers for selecting elite long-distance runners.The case-control studies were used to analyze distribution characteristic of the gene polymorphisms between 123 elite long-distance runners and 134 controls. These polymorphisms were located in HEPC (rs7251432), FPN1 (rs1123110,rs4145237, rs11568350'rs11568351)andDMT1(rs12227734, rs224589, rs149411,rs1632791、rs61078145 and STR repeat polymorphism) gene.The results showed the frequencies of CG genotype of Master Sportsman group was significantly higher than those in control group for rs4145237, and frequencies of CC haplotype in FPN1 with both Master sportsman and marathon group were significantly higher than those in control group. Moreover, the frequencies of ACC haplotype and homozygous 258bp of STR polymorphism in DMT1 with athlete groups were significantly higher than those in controls. To indicat CG genotype of rs4145237 and haplotype of FPN1, ACC genotype and homozygous 258bp of STR polymorphism of DMT1 can be used as a genetic marker in selecting elite long-distance runners. However, rs11568350, rs11568351, rs1632791 and rs61078145 in northern Chinese Han population not showed the polymorphism.Part II:The feasibility of HEPC、FPN1 and other genes as genetic markers for predicting the response to aerobic training.This study explored the association between these gene polymorphisms and aerobic performance parameters at baseline and its response to 18-week aerobic training in 102 males from Northern China (Han origin) by adopting association studies with training response phenotypes methods.The results showed the increment of 150W/SV and 150W/SVI with AG genotype in rs7251432 were significantly higher than those none carrier, and increment of 150W/EF was significantly higher then GG and AA genotype too.The increment of 100W/SV and 100W/SVI with CC genotype in rs1123110 were significantly higher than TT genotype, and decline in the amount of CO and COI were significantly higher than TT and TC genotype. The increment of 100W/EF, 150W/T, and decline in the amount of Re/CO, Re/COI with AG genotype in rs 12227734 were significantly higher than GG genotype, and increment of Re/T was significantly higher than AA and GG genotype. The incipient with ESD、EDD and CO were highest for GG genotype in rs4145237, but the increment of PWS and Re/EF were highest. To indicated AG genotype of rs7251432, CC genotype of rs1123110, CG genotype of rs4145237 and AG genotype of rs12227734 would be used as genetic markers of the response to left ventricular structure and/or function to aerobic training. None association was showed between gene polymorphism above mentioned and both VO2max and RE, prompting those polymorphisms can not been used as genetic markers of VO2max and RE.Part III:The feasibility of HEPC、FPN1 and other genes polymorphisms as genetic markers of predicting the response to HiHiLo trainingThis study analyzed the association between these gene polymorphisms and aerobic performance parameters at baseline and its response to 30-day HiHiLo training in 71 healthy young males with Plain living from Northern China by adopting association studies with training response phenotypes methods.The results showed the increment of PWD and IVSD with AA genotype in rs7251432 were significantly higher than AG genotype, and decline in the amount of 100W/HR was significantly higher than GG genotype. The increment of Re/SV and Re/SVI with AA genotype in rs7251432 was significantly higher than AG and GG genotype. The incipient of LVM and LVMI with CC genotype was significantly higher than AC genotype in rs224589. Decline in the amount of Re/HR with 254/262bp genotype of STR was significantly higher than other genotypes. The increment of RBC, Hb and Hct with AA genotype in rs7251432, TT genotype in rs1123110,GG genotype in rs4145237 and 254/258bp genotype in STR were significantly higher than those none carrier after training. Therefore AA genotype in rs7251432, CC genotype in rs224589 and 254/262bp genotype in STR would be used as genetic markers of the response to left ventricular structure and/or function to HiHiLo training. Moreover AA genotype in rs7251432, TT genotype in rs1123110, GG genotype in rs4145237 and 254/258bp genotype in STR could be used as the genetic markers of the response to Hematology Parameters. None association was showed between gene polymorphisms above mentioned and VO2max and SpO2, prompting those can not been used as genetic marker of VO2max and SpO2.
