Purpose:The aim of this work was to investigate the association between aerobic capacity performance?both baseline and training response?in general population of Han Chinese and AGTR1 gene polymorphism,to know whether AGTR1 gene can be able to predict individual differences of aerobic capacity performance?both baseline and training response?as an genetic marker.Subjects:1)963 Ordinary people?who had not participated in the regular physical exercise within 1 year?as baseline subjects.2)511 people?of the 963 subjects who completed 16 weeks exercise training?as training response subjects.)Methods:9 tag SNPs?rs12721241,rs12721276,rs6801836,rs275652,rs5182,rs3772616,rs385338,rs2131127,rs2675511?of AGTR1 gene were selected via haploview software and Hapmap database.MALDI-TOF MS technology was used to examine the genotype.Physique indicators,body composition,aerobic endurance,cardiac structure and function indicators?both baseline and training response?were measured to analyze the association between these indicators and AGTR1 gene polymorphism.SNPs polymorphism preliminary functional analysis was done via online software after the associated loci was found.Results:1)The distribution of Genotypes on rs6801836,rs275652,rs385338,rs2131127,rs5182 that in line with the Hardy-Weinberg equilibrium,suggests the selected group is qualified as the representative of the population.2)The distribution of AGTR1 gene polymorphism has shown a significant racial difference.3)In women,the VO2max and VO2max-MET of the CC genotype in rs275652 were significantly lower than those in AA and AC genotype.In both genders,the percentage of region fat,fat mass of body composition and skin fold thickness on multiple sites the of CC genotype were significantly higher than those in AA,AC genotype.Functional analysis'results showed that the loci with A/C transversion cause no functional differences.4)In men,the LVDd,LVEDV,SV of CC genotype in rs6801836 was significantly lower after training compared to AA,AC genotype.The reduction of Diastolic blood pressure of CT genotype after training was significantly greater than the CC,TT genotype in both genders.Functional analysis'results showed that the loci with C/T transversion cause no functional differences.5)In women,the reduction of skin fold thickness on iliac and thigh of rs2131127 TT genotype after training was significantly greater than those in CC and CT genotype.Functional analysis'results showed that the loci with C/T transversion cause no functional differences.Conclusion:1)CC genotype in rs275652 may mark a poor initial aerobic capacity.2)CC genotype in rs6801836 showed poor sensitivity on left ventricular structure index after training in male subjects.3)TT genotype in rs2131127may have a high sensitivity on fat content reduce after training in women.
