In this study, the whole process of micropropagation of gerbera in vitro was conducted. The main results are as follows.There lay differences in callus induction of leaf on the various media. Callus was induced on MS medium, but not on other media. Among leaf, floweret, floral receptacle and floral peduncle, 2,4-D and 6-BA had better effect on induction of callus than other auxins and cytokinins. Tndse four explants could all be induced to produce callus, of which the induction rate of floral peduncle was the highest. It reached 96.8%. Leaf and receptacle had the highest induction rate of callus on the media of MS +6-BA2.5 mg.L-1 +2,4-D 0.5mg.L-1+NAA0.4 mg.L-1 and MS+6-BA2.5mg.L4+2,4-D1.0 mg.L-1 +NAA0.2mg.L-1, respectively. Adventitious buds could be obtained from these calli quite well.Soaking leaf explant in the solution of 100mg.L-1Vc before being planted and adding 200mg.L-1 PVP to medium during subculture could control browning very well.Variant explants showed differences in differentiation. Adventitious buds were induced directly from floral receptacle, but not from leaf. There were different effects on differentiation of adventitious buds among cytokinins. 5.0mg.L-16-BA had the best effect on differentiation of adventitious buds. The effect of KT was slightly less than that of 6-BA. Lower concentrations of auxins were necessary to the induction of adventitious buds. 0.2mg.L-1NAA was optimal. Although higher concentrations of auxins were not suitable for the induction of adventitious buds, they were good for elongation of shoots.Combinations of different ingredients in medium exhibited significant effect on the proliferation of adventitious bud. Combinations of 6-BA3mg.L-1, NAA0.3mg.L-1 with CC3mg.L-1 and GA0.5mg.L-1, sucrose40g.L-1 with agar6g.L-1 had the best effects on the proliferation and development of adventitious bud.1/2MS basic medium was better than MS and 1/4MS media for root induction. Root induction was promoted greatly in 1/2 MS medium supplemented with 0.05-0.1 mg.L-1NAA or 0.2-0.3% activated charcoal. Transplanting rooted shoots to substrate composed withlperlite: Ipeat, survival rate of shoots was increased to 95.4%. At the same time, the regenerated plantlets grew vigorously after transplant.
