Light source is one of the most important factor for Tissue Culture,it is not only provide radiant energy for plant photosynthesis,but also provide signals for the plant to adjust its development.The purpose of this experiment is to get high-quality in vitro and large-scale production, The use of fixed-wavelength, light intensity adjustable light-emitting diodes and the cold cathode fluorescent lamp as a light source.Use Gerbera"Ruikoufor"the experiment plant material to explore the effectiveness of light-emitting diodes and the cold cathode fluorescent lamp in the production for Gerbera jamesonii in vitro,and also comparison with ordinary fluorescent lamp to Gerbera factory for costing.1. Application of light-emitting diode (LED) as light source for in vitro culture of Gerbera jamesonii.The results showed that light-emitting diode light source make a better role in promoting the growth of Gerbera jamesonii in vitro and the growth of transplanted than conventional fluorescent lamp. Gerbera varieties"Ruikou"in the experiment,In the 50μmol ? m-2 ? s-1 under light intensity of the red / blue ratio of 5:5, under conditions of light quality growth best,Plant height, leaf number, leaf length, leaf span reached 57.2 mm, 8.1 , 18.1 mm, 14.8 mm,Fresh weight for the whole 641.4 mg, dry weight for the whole 64.3 mg.2. Applications Cold Cathode Fluorescent Lamp (CCFL) as a light source for in vitro culture of Gerbera jamesonii. The results showed that: Cold Cathode Fluorescent Lamp make a better role in promoting the growth of Gerbera jamesonii in vitro and the growth of transplanted than conventional fluorescent lamp. Gerbera varieties'Ruikou'in the experiment,In the 30μmol ? m-2 ? s-1 under light intensity of the red / blue ratio of 7:3, under conditions of light quality growth best,Plant height, leaf number, leaf length, leaf span, root length reached 48.5 mm, 8.0 , 16.0 mm, 15.0 mm,29.1mm, Fresh weight for the whole 641.4 mg, dry weight for the whole 64.3 mg.3.The use of LED and CCFL light source,tissue culture rooms decreased power consumption. A CCFL light source as an example, the production of one million test-tube seedlings Gerbera power consumption cost of 0.105 kwh / bottle of (5 / bottle), the total annual electricity consumption for the 26 443.2 kwh, electricity costs 26 443.2 yuan ( 1 yuan in industrial electricity / kwh basis), it can reduce costs each year nearly 60,000 yuan than the use of fluorescent lamps. The results show that the light-emitting diodes, cold cathode fluorescent lamp used in in vitro culture of Gerbera jamesonii can significantly improve the in vitro and the growth of transplanted indicators. The results will have a positive impact on the further development of Gerbera jamesonii in vitro.
