Oil-tea is the main woody edible oil tree in the South of China. It plays a very important role in people's daily life and our national economy; however, its output is low at present. Haploid breeding is advantageous in many ways in tree breeding. In order to solve the problem of low production, we tried to improve oiltea varieties by applying the haploid breeding technology. Because very few studies have been done on oil-tea anther culture, this study was focused on the basic researches of the haploid breeding.1 Induction of oil-tea anther calliThe study aimed at raising the frequency of induction anther calli.Eight oil-tea species were used as test mateiral, namely, Camellia oleifera Abel., C. meiocarpa Hu., C. yuhsienensis Hu., C. chekiangoleosa Hu., C. semiserrata Chi., C. reticulata Lindl., C. vietnamensis Huang., and C. polyodonta How.ex Hu. Through a series of experiments, the establishment of technical system for oil-tea haploid breeding has attained staged progress.After the formation and morphology of bud and anther were found in the stage of single nuclear microspore by observation, in the process of anther calli induction, the basic method of oil-tea anther culture was developed, and the technical scheme of inducing oil-tea anther calli was preliminarily selected. Experiment results show:(1) Of the eight oil-tea species, the frequency of induction anther calli from Camellia oleifera Abel, and C. meiocarpa Hu. were higher than others.(2) Pretreatment: The pretreatment of low temperature was helpful for inducing oil-tea anther calli, and the handling time differed with the species. Centrifugal pretreatment could also raise the frequency of induction.(3) Hormone: Under 2,4-D 0.5mg/L and 6-BA 0.2mg/L the effect of induction was better in the experiment, in which 6-BA was the leading factor. In the hormone combination of 2,4-D and NAA, 2,4-D played a leading role, the frequency of induction raised when the concentration of 2,4-D increased.(4) Culture medium: MS culture medium was fit for oil-tea anther culture.
