The genetic diversity of six goose breeds (White Goose, Zi Goose, Huoyan Goose, Wanxi Goose,Rhin, Landoise) was analyzed by microsatellite markers. The genetic variation of population(includingpolymorphism information content, gene frequency of alleles, heterozygosity and effective number ofalleles et al) of 6 microsatellite loci was analyzed by using DNA extraction, PCR amplifying,polyacrylamide gel electrophoresis testing et al.The results revealed that seven microsatellite loci in six breeds have different genetic variation. Allloci showed high polymorphism. There were 70 alleles inspected in 7 microsatellite loci, the averagenumber of alleles was 7, number of alleles ranged from 6~16, Allele frequencies were 0.0185~0.7000,6 goose breeds had 7 the same alleles. The alleles distribution of each locus was different among breeds,and the dominant alleles'frequency was also different between breeds. There were 16 alleles in WD206,which was most, and the least was WD136 which had 6 alleles.The range of mean PIC was 0.5497~0.7766, and mean heterozygosity in every population rangedfrom 0.6617~0.8134, it was highest in Zi goose and lowest in Rhin. The significantly higherheterozygosity suggested that the native breeds have abundant genetic diversity and potential ofselection.Using the polymorphism information by 7 microsatellite loci providing, Genetic distance could becalculated between different breeds. The UPGMA cluster analysis put the White Goose, Zi Goose,Huoyan Goose and Wanxi Goose into one group, and the foreign breeds of Rhin and Landoise intoanother.The result of UPGMA is consistent with genesis, geographical distribution and economic typeof these breeds.The dendrogram obtains from genetic distance could correctly reflect the phylogeneticrelationship among the six goose breeds, suggest that microsatellite DNA marker is a useful tool todetermine genetic diversity in closely related breeds.
