Over time,Molecular markers have proven to be fast and dependable apparatus of classifying mushroom population.Here are determine the genomic diversity among five species of JUNCAO Pleurotus:Pleurotus Ostratus "P969",Pleurotus djamor "Red",Pleurotus Cap Fungi "PL-30",Pleurotus eryngii "Yellow",Pleurotus geesteranus "P.G.they were characterized using ITS and RAPD,the ITS region of rDNA were amplified using PCR.Finding shows that the size of the ITS from the different strains varied between 680 to 1300 bp,respectively,the ITS band was polymorphic in all the strains.A phylogenetic tree based on the ITS region indicated that selected strains could be classified into three clusters.Among the fourteen random amplified polymorphic DNA(RAPD)primers used DTT 10 and DTT 11 had the good polymorphic and securable band pattern;while the polymorphic band of DTT-9 observed was not completely visible in one of the specie(Pleurotus cap fungi(PL-30).To estimate the homology and genomic distance among the different species,cluster analysis was used based on frequency similarity,the genomic relations among species ranged from 50%to 95%,and the similarity among three species of cluster A,(Yellow,P.G.and PL-30)was from 80%to 95%,cluster‘B'comprised of two species(P969 and Red);with 65%similarity.The similarity matrix shows that most correlated species were(Yellow and PL-30),with 95%homology.A further study was done to demonstrate the effectiveness of using JUNCAO grass in cultivating the five strains of Pleurotus and determined the most efficient formula,the grasses were 1)Pennisetum purpurum(Giant JUNCAO),2)Dicranoteris dichotoma(Elephant),3)Lesotho,4)Vetiveria zizantiodes,5)Mischantum Floridulus,investigation was done on their growth and yield parameters.Result indicated that Pleurotus Ostratus"P969" showed the highest biological yield and efficiency than other strain in all the formulas,also,the mycelium run rate and number of fruiting bodies were higher in Pleurotus Ostratus "P969" and Pleurotus djamor"Red" than other three strains.The quality of mushroom strains was almost similar.Our study have shown that,JUNCAO Technology could be used as novel,practical,and easily accessible alternative substrates for Pleurotus mushroom cultivation,it also demonstrated the effectiveness in using ITS and RAPD markers in genetic diversity,characterization and differentiation of Pleurotus mushroom cultivars.
