Flavonoid is widely researched in nature product exploitation because of its specific activity to human body. Up to now, there is none of report on the purification or determination of bee-pollen total flavonoids. The present studies took an investigation into these fields, and established a set of methods, which are economic and safe for purification and quantitative determination of bee-pollen total flavonoids.The present studies developed a method for quantitative determination of the total content of bee-pollen flavonoids via ployamide-UV spectrophotometry. The contrast sample was rutin for ployamide-UV spectrophotometry. The optimum determination wavelength was 360nm. The result showed that a good linearity relationship (correlation coefficient: 0.9998) between the degree of absorbance and the concentration of analyte was obtained, within the range of 0.005—0.025mg/ml. The average recovery rates were 98.7%, the measurement precision was high with its RSD 0.488%(n=3).The present studies implemented ployamide to enrich bee-pollen total flavonoids. Static adsorption test and static de-adsorption test were carried out to indicate that ployamide possessed strong adsorption ability to bee-pollen total flavonoids, in addition of an easy de-adsorption property. Dynamic adsorption test and dynamic de-adsorption test were carried out to make sure the condition of purification in bee-pollen total flavonoids. The dynamic adsorption amount was 42.44mg/g and de-adsorption ratio was about 96%. The content of total flavonoids of finally product was amount to 20%, enhanced 13 times than that of raw material.
