The officinal plant Gynostemma pentaphyllum (Thunb) Makino is one kind of perennial herb liane, which belongs to Cucurbitaceae and the whole plant is officinal. There is a long history in China that people have planted it for strengthen body and treatment. Many Chinese traditional documents such as "Compendium of Materia Medica" "Textual Research on Reality and Titles of Plants" etc. have the record thatG pentaphyllum is a traditional medicine. The pharmacology research indicate that G pentaphyllum has a wide curative effects as behaving antixidation, diminishing inflammation, relieving a cough , detoxification and dispelling phlegm, curing tracheitis and contagious hepatitis, promoting immunity, calming, leading hypnogenesis, reducing fat in the blood etc. Additionally, G pentaphyllum is mainly produced in the southern part of China and then has a famous name called"south Panax" .G pentaphyllum plants are distributing widely in China and there exist a large of differences between them. Carry through the study of its resources can be the basis of other research such as selecting the good breed, developing the genetic theory and biotechnology. Establish the uniform descriptors and data standard is very pivotal and have many advantages. It is propitious to conforming the whole resources of our country, making criterion to the collection and conservation of G pentaphyllum and creating a favourable environment to the sharing of all the information.The establishment of the data standard can also in favor of the proper resources using of G pentaphyllum. After the collecting and investigation research, the develop of G pentaphyllum may enter into a more well-ordered and high-efficiency way and then can accelerate the speed of its development to show its full value to society.Based on the research of investigation and collection in the main area inChina , we evaluated the different idioplasmic resources of G pentaphyllum according to its morphology and quality characters. In addition, we established its DNA fingerprint by molecular markers and ultimately constituted the descriptors and data standard of different idioplasmic resources of G pentaphyllum. The main results of this research could be concluded as folio wings:1) We have led an in-depth and detailed investigation of the wild G pentaphyllum as well as the cultivated in the main regions where produce G pentaphyllum in China. The investigation included many factors such as geography factors, entironment, peculiarities and so on. Based on the investigation we summarized the actuality of the using condition and then pointed out the problems existing in the using of G pentaphyllum. More or less, we have put forward the referrence counter measures to solve the problems.2) The paper studied on the diversities of different G pentaphyllum from all morphology characters including root, stem, leaf, flower, fruit and seed. A relative systematic statistic analysis was then carried out to discuss the diversity and even the genetic variety of all those resources of G pentaphyllum. The statistic results indicated that 5 characters among 12 differed apparently—P<0.05> and in these 5 characters, the length/width of leaf, leaf hair and seed size differed most apparently —PO.01. Therefore, these characters can be considered to be the most important characters in identifying the differentiation of the different resources of G pentaphyllum3) From the comparative results of the quality character of the different G pentaphyllum, we evaluated all resources systematically and further put up some good breeds as the favourable plants in subsequent selective cultivation.4) DNA fingerprint by RAPD analysis were figured out in this paper. The study of G pentaphyllum was based on the optimized RAPD analysis which had different genomic DNA as templates in PCR amplification. The optical parameters in 20uL PCR reaction system including 10Xbuffer2.0uL, Mg2+2mM, dNTP (UmmolL'1, random primer 0. SmmolL"1, Taq DNA polymerase 1U, and template DNA 50ng and reacted in the optimized protocol: 94°C3min— (94°C30 s -?38°C40 s -*72"C lmin) 45cycles-*72°C10min-i-4<>Ckeepingo Five primers were selected from 66random ones to perform this RAPD amplification. The results helped to locate the characteristic polymorphous sites of Gpentaphyllum as well as establish the identification key based on molecular markers.5) According to the relative principles and rules, we have constituted the descriptors and data standard of different idioplasmic resources of G pentaphyllum. The final standard database is huge and involved geographical character of distribution, morphological character, quality character and character in molecular markers etc.
