The plant Gynostemma Pentophyllum (Thunb.) Makino is a kind of Perennial herb liane, which belongs to Cucurbitaceae. Except gypenosides flavonodids are the major medicinal components that are extracted form the leaf, stem, rhizome and root of this herb.1. The optimum conditions of extraction process for rutin from Gynostemma Pentaphyllum were selected with extraction phase, the concentration and extraction of time by orthogonal experimental design. The optimum preparative procedure was as follows: Gynostemma Pentaphyllum was extracted with ultrasonic technology 60 min at 70% methanol. The method is simple, stable, accurate, and reproducible and provides a scientific basis for improving the rate of extraction of rutin from Gynostemma.2. We study on the High Performance Liquid Chromatography (HPLC) Fingerprint of the Gynostemma Pentaphyllum. We study the methods of the sample solution preparation, the selection of reference matter, the optimization of Chromatography factors (i.e. the selection of Chromatography column, fluid phase, detecting wave). The HPLC-fingerprint of Gynostemma Pentaphyllum was established. We test precision, repeatability and stability of experiment. So the method is reliable.3. We establish the fingerprint spectrum of flavonoid from Gynostemma Pentaphyllum. by using the HPLC conditions. Ten common characteristic peaks were taken as fingerprint peaks the precision and the retest are in accordance with the requirement. The method is stable, reliable and precise. It provides a scientific basis for the quality standard of Gynostemma Pentaphyllum.4. Append different concentration Methyl Jasmonate to MS culture medium, can enhance the total flavonoid content in callus of Gynostemma. Low concentration of MJ can increase the total flavonoid contents and the activity of PAL,POD,SOD and CAT in callus of Gynostemma. The deal whit 50μmol/L MJ have significant correlation whit antitheses. Those maybe indicate that after deal with MJ, series of enzymes genes are induced to depress, and soon the plant secondary metabolites are enhanced, the total flavonoid contents are increased.
