Low-energy ion implantation is utilized in this research. We use the different implantation dosage to inject into cell of the Pleurotus ferulae Lanzi. Trained by the solid and the liquid ferments, after the repeated implantation and filter,we got the best implantation dosage:9×1014N+/cm2. The strain GC-1 was screened out according to the yield of extracellular polysaccharide.The amount of polysaccharide,which reached 4.75g/L,is increased by 35% compared to strain without ion implantation. In this research we Optimize the Liquid training for Pleurotus ferulae Lanzi GC-1,and comfirm the best culture medium:the powder of wheat 2%, sugarcane 2%,yeast extract 0.4%,peptone 0.2%, KH2PO4 0.2%,MgSO4 0.1%,bran 3%.PH 7.0, 30℃,200r/min,1/5 medium volume.The best time of inoculate is 60h,the measure of inoculate is 10%.we olse optimumed the techincs of the isolation and purification Of polysaccharides,and get the best scheme of immerses :temperature 100℃, immerse two times,the ratio of dimension is 20:1,the times of immerse is 3h, the best scheme of removing protein : Sample/Trichloromethane-Butyl alcohol(V/V) is 1/1, Trichloromethane/ Butylalcohol(V/V) 2/1,time 40min;75% concentration of ethanol pepositing.
