Both Magnolia officinalis Rehd. et Wils. and Magnolia biloba (Rdhd.et wils.) Cheng. are members of Magnolia genus of Magnoliaceae familiy. With the properties such as warm-natured and bitter taste, they were used for the treatment of pneumatosis, constipation, dyspnea with cough, vomiting and symposiums diarrhea as a traditional Chinese medicine for hundreds years.The chemical constituents such as of magolol, honokiol and magnocurarine compounds were the major contents in Magnolia officinalis, which possessed the effect on flaccidity, anti-anabrosis, central inhibition, lower blood pressure and anti-tumor in modern pharmacological research. According to the Pharmacopoeia of China (Ch. P. 2005 Edition), the content of hydroxybenzene derivatives in the ideal material Magnolia officinalis should be up to 2%. In order to find a quick and accurate method to assay magnolol and honokiol, three extraction methods were used to value the magnolol and honokiol, which were extracted by methanol, with ethanol and CO2 supercritical fluid extraction. The result showed that CO2 supercritical fluid extraction (CO2 SFE) could be the most efficient. The content of magnolol and honokiol were assayed by HPLC anlysis. According to the quantity test values, the result of total concentration of magonlol and honokiol was up to 47.9% showed that the method of CO2 SFE was superior to the traditional methods with solvents.In order to find out the chemical constituents of CO2 SFE extract, the study was carried out, by means of the silica gel column chromatography, Sephadex LH-20 column chromatography and other isolation methods, six compounds were obtained and their structures were elucidated as Magnolol(l), Honokiol(2), Magnaldehyde C(3), Magnaldehyde B(4), Crytomeridiol (5) andβ-Sitosterol (6), respectively. Among them, compound 5 was isolated from this plant for the first time.
