The stem bark of M.officinalis(magnoliae officinalis cortex)has been extensively used as a traditional Chinese medicine for treatments of abdominal distention and pain,dyspepsia,and phlegm of cough with asthma.As a traditional Chinese medicine aromatic,previous chemical studies focused on its lipophilic ingredients.The study of water-soluble ingredients are rare.Traditionally,magnoliae officinalis cortex is boiled in water to extract active ingredients hydrophilic ingredients were thus regarded to have more relation with their clinic effect.However,little work has been done on the hydrophilic ingredients.Magnolia bark is an important drug in the treatment of metabolic disease,magnolia bark and previous studies have shown that the total phenol has a certain degree of hypoglycemic effect.In this paper,through the study of the chemical composition of water-soluble part,further clarify the mechanism of magnolia officinalis to regulate blood sugar metabolism.To study the water-soluble chemical constituents from the Stem Bark of Magnolia officinalis,the chemical constituents were isolated and purified by silica gel,ODS column chromatography,Sephadex LH-20 and semi-preparing HPLC.Their structures were identified on the basis of ESI-MS,UV and NMR spectra.As a result,nineteen compounds were isolated and elucidated which included 13 phenylpropanoids:(7S,8R)-syringoylglycerol(1),3,4-dihydroxy-allylbenzene-3-O-L · rhamnopyranosyl(1?2)?-D-glucopyranoside(2),3,4-dihydroxy-allylbenzene-3-O-L-rhamnopyranosyl(1?6)?-D-glucopyranoside(3),synapticaldehyde-4-O-?-D-glucopyranoside(4),syringaresinol-di-O-?-D-glucopynanoside(5),Magnolol(7),Honokiol(8),4-O-methylhonokiol(9),syringing(10),acteoside(11),Magnolignan C(13),syringaresinol-4-O-?-D-glucopyranoside(17),3,3',9'-trimethoxy-4',5,9 · trihydroxy-5',8-bilign-7'-ene-4'-O-?-D-glucopyranoside(19),3 flavonoids:Luteolin-7-O-?-D-gulcoside(12),liquiritin(14),liquiritigenin-7-O-?-D-gulcoside(15),3,4,5-trimethoxyphenyl-?-D-glucopyranoside(6),2,4,6-trimethoxyphenol-1-O-D-glucopyranoside(16),Octadec-6-enoic acid 2,5-dihydroxy-4-octanoyloxy-phenyl ester(18).Compounds 6,12,14?16 were isolated from this plant for the first time.Compound 19 was a new compound,and has not been reported before.Four compounds with hydroxyl of the water-soluble part of Magnolia phenylpropanoid structure were selected.High glucose cultured HepG2 cells,1,10,100,200 ?M after treating cells,MTT method was used to examine the water soluble components 3,4-dihydroxy-allylbenzene-3-O-L rhamnopyranosyl(1?2)?-D-glucopyranoside,syringaresinol-di-O-?-D-glucopynanoside,syringing,3,4,5-trimethoxyphenyl-?-D-glucopyranoside in cultured human HepG2 cells of high glucose.Insulin sulfuric acid anthrone colorimetric method to stimulate cell glycogen synthesis was used to detect.The results show that four kinds of drugs can significantly improve insulin sensitivity in the cell,especially in the 100 ?M syringoside HepG2 cells particularly significant effect,achieve the effect of the 89.37%Rosiglitazone Hydrochloride positive control group.Due to the liver for sugar in the body storage,decomposition and regulation of blood glucose and other aspects are crucial,and with the phenolic hydroxyl group of compounds can reduce free radicals in the body,from the oxidation of free radicals on the liver cell membrane lipid and protein,protect the structure and function of liver cells can restore the normal function of.It is speculated that this important drug honokiol in treatment of metabolic diseases may improve liver function to achieve the purpose of treatment of diabetes.
