Gene flow is the transfer of alleles of genes from one population to another. According to the acreage of crops,more than half of China's Bt transgenic cotton used for mushroom cultivation. Among these,lots of them used to cultivate mushroom. Foreign gene of transgenic insect-resistant cotton flow to the outside. Therefore,it is very important to study the fate of Bt gene.1.The cultures containing MON15985 were used to cultivate P. ostreatus Understanding the composition of genetically modified cottonseed. The results showed there was no significant difference in yield and besides other characteristics of mushroom. It indicated transgentic cotton seeds could be used as ingredients.2.Comparing of the CTAB method,SDS-CTAB extraction method and extraction method with magnetic beads,measuring the quality of the genome by UV spectrophotometer,restriction enzyme digestion and PCR tests. The results showed that the SDS-CTAB method and extraction method with magnetic bead meet operational requirements,the former lower cost while the yield higher.3. Considering the cost,we decide to choose SDS-CTAB method to continue the research. Explored the fragments of foreign DNA by PCR,the mushroom spawn several generations could be detected the fragments of CaMV35S promoter,NOS terminator and Sad internal standard gene cottonseed,Bt gene fragment. So the cells of mushroom acceptted foreign DNA during the mushroom cultivation process.4. Extracting RNA of mushroon fruiting body and detecting whether there was the possibility of genetic drift by RT-PCR. The results showed that the mushroom don't accept any foreign fragments at the transcriptional level.
