Polygonatum sibiricum Red. is a traditional chinese herbal medicine which belongs to the Liliaceae family and is widely distributed in southwest of China. It has various pharmacological functions, for example, anti-senility, anti-tumor, hypoxia-tolerance, and so on. The major chemical compounds in P.sibiricum Red were polysaccharides, steroidal saponins, flavonoids and anthraquinones.At present, there are many kinds of stems and roots of the wild genus in Liliaceae family appeared as medicine rescourses in the market . It was difficult to keep the herbal medicine under the quality control, for the harvesting and the processing procedure mainly depending on the traditional experiences. In rcent years, with the development of modernization of traditional chinese medicine and the GAP bases being established, the stantardization cultivation and processing of P.sibiricum Red. were carried on gradually. Hanzhong region is the main production area in china. So, in the present paper, the duration of cultivation, the harvesting time and the processing methods of Huang-Jing from HanZhong GAP base were studied. The accumulation and distribution of sacchrides in the stems were analysed, the influence of the different processing methods on the quality were investigated. Based on the results above, some compounds were separated and purified from the ethanol extractsfrom the stem of P. sibiricum Red by modern chemical isolation and purification methods. And the structures of them were elucidated by the physicochemical properties detection and a series of spectral analysis. After these researches, the main purposes are to investigate the dry matter and polaysaacharides accumulation and distribution traits in order to determine the best harvesting time and the properly processing method and provide some technical informations for Huanjin GAP base development.The experimental results were achieved as follows:1. Using the anthrone-sulphuric acid colorimetry, the sacchride contents in the samples of P.sibiricum Red. were determined in order to investigate its duration of cultivation. The results showed that the sacchride contents in P.sibiricum Red were quite different between different age sections. The contents were higher in one, two and three years section, 4.48%, 4.39%, 4.45% respectively. It got lowered later and for the six-year section, was the lowest, about 3.50%. As the growth year last longer, the water extracts and the sacchrides content decreased apparently, on the contrary the alcohol extracts increased apparently. Considering...
