Study Of Prevention-cure Effect And Mechanism Of The Complex Perfusion Fluid Of Traditional Chinese Medicine For Dairy Cow Mastitis

The mastitis is an inflammatory reaction of mammary gland of dairy cow caused by the stimulation, which can lead to loss lactating function. It will take serious economic losses to dairy farm. According to the theory of syndrome differentiation and treatment of traditional Chinese veterinary medicine and the pathological features of mastitis, in combination with modern injection preparation techniques, the complex perfusion fluid of traditional Chinese medicine for treating mastitis was triturated and selected, which was composed of japanese milkwort herb, Herba Gueldenstaedtiae, Barbated Skullcup Herb, Tokyo violet herb, and so on. The complex perfusion fluid of traditional Chinese medicine had the effects of clearing heat and detoxicating, bacteriostasis and dephlogistication, detumescence and odynolysis, promoting blood flow and dredging the meridian passage. In order to study the mechanism, drug action and clinical safety of the complex perfusion fluid of traditional Chinese medicine, the drug action, the perfusion safety test and clinical prevention and cure effect were also investigated.Pharmacodynamic experiment: Bacteriostasis activity in vitro was observed by agar diffusion and tube double dilution, the analgesic effect was observed by Woolfe-Macdonald method and acetic acid stimulation inducing writhing action model in mouse, the anti-inflammatory effect was observed by acetic acid inducing penetration increase of mouse abdominal blood capillary model and dimethylbenzene inducing ear edema model, the relieve fever effect was observed by fever response model of rabbits induced by Typhoid-paratyphoid vaccine, immunologic enhancement effect was observed by immune organs and peritoneal macrophage phagocytic ability of mouse fed with complex perfusion fluid continuity. The results showed that the complex perfusion fluid of traditional Chinese medicine had well bacteriostasis on Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli and Candida tropicalis, increased the percentage of pain threshold and resisted the writhing action caused by acetic acid significantly in mouse (p<0.01), an significant inhibitory action on acetic acid inducing mouse abdominal capillary permeability (p<0.05or p<0.01), an significant resistant effect on dimethylbenzene inducing ear edema model (p<0.01), significant antipyretic effect on Typhoid-paratyphoid vaccine inducing fever model (p<0.05or p<0.01), the weight of immune organs and splenic corpuscle increased significantly, phagocytic activity of peritoneal macrophage of mouse enhanced significantly (p<0.01or<0.05).Safety experiment: The acute toxicity, haemolyticus, irritation and sensitization of the complex perfusion fluid of traditional Chinese medicine to dairy cow breast were infused in breast. These influences were observed by the acute toxicity test on mice, hemolysis and agglutination test, stimulation test on intramuscular of rabbits and breast of sheep perfusion with the complex perfusion fluid of traditional Chinese medicine, pyrogen test and sensitivity test in health guinea pigs. The results showed that the complex perfusion fluid of traditional Chinese medicine had no acute toxicity in mouse, no haemolyticus and irritation, and no sensitization to rabbits and guinea pigs. It showed that the complex perfusion fluid of traditional Chinese medicine can be used to infuse in breast.Clinical experiment: On bases of bacteriostasis activity, anti-inflammatory experiment, antipyretic analgesic and immune-enhancement experiment and security test, the complex perfusion fluid II of traditional Chinese medicine was determined to prevention-cure mastitis in dairy cows.40dairy cows with mastitis and40healthy cows were selected for prevention-cure and immune test in clinic. The somatic cells count in milk, SA and the enzyme levels in the whey and SI and the rate of neutrophil phagocytosis during cure were measured. The results showed that the cure rates of sub-clinical mastitis by the complex perfusion fluid of traditional Chinese medicine and antibiotics were respectively94.44%and88.24%, and the effective rates were100%. The cure rates of clinical mastitis in dairy cows were respectively86.67%and84.62%, and the effective rates were93.33%and91.67%. The preventive effect of mastitis in dry period was88.89%. Comparied with control group, the levels of somatic cells count, SA, AST, ALT and LDH were increased significantly before treatment (p<0.05or p<0.01) the complex perfusion fluid of traditional Chinese medicine treatment5-7d, the above-mentioned indexes were reduced to normal levels and not significant compared with healthy cows (p>0.05), but only LDH reduced to the normal level in subclinical mastitis group treatment with antibiotic, and the levels of SI and the rate of neutrophil phagocytosis were increased significantly (p<0.01).The experiments confirmed that the complex perfusion fluid of traditional Chinese medicine had a reliability treatment effect on subclinical and clinical mastitis, and could reduce the number of somatic cells count and SA and enzyme levels in the whey, improve SI and the rate of neutrophil phagocytosis. The experiment showed that the complex perfusion fluid of traditional Chinese medicine could repair damaged mammary gland epithelial cells, improve the immune function of dairy cows, and advance rehabilitation of the ill cows.In conclusion, the complex perfusion fluid of traditional Chinese medicine had obvious pharmacodynamic effect of bacteriostasis, anti-inflammatory, antipyretic analgesic and immune-enhancement, and can be used to infuse safely in breast. It had a good effect in clinical prevention and cure, could repair damaged mammary gland epithelial cells, and improve immune function.
