Polysaccharide,the richest member of biologic macromolecule,is the natural macromolecule compond which is making up of monosaccharide and almost exist in every organism, including animals, plants and microbes. The polysaccharide of Pleurotus ferulae Lenzi play an important role in antitumor, antivirus and degrading glycemia, it is not only an ideal medicine, but also a functional ingredient of health food, so the foreground is wider. At the present time, the researches of this polysaccharide are very few, so the key problem of developing is to increase the content of polysaccharide.In order to increase the content of mycelium polysaccharide, mycelium mono-cells of Pleurotus ferulae Lenzi were treated tentatively by ion beam implantation. Two polysaccharide high-yield strains, PFPH-2 and PFPH-3, were selected through fermentation breeding after primary breeding. The polysaccharide yield of these mutants were 551.80mg/L and 659.46mg/L separately, which were increased by 46.5% and 75.2% compared to that of initial strain. Then PFPH-2and PFPH-3 used as the original strains were mutated by ultraviolet light and LiCl. The results indicated that the polysaccharide yield of strains 1, 9 and 10 were decreased by 27%, 38% and 37% compared to that of PFPH-2 and the polysaccharide yield of strain 17 was decreased by 28% comparied to that of PFPH-3 through fermentation quantitative breeding after primary breeding.
