Sedirea japonica belongs to Orchidaceae , Sedirea , and is the national two level of protection plants. More and more people love it because of its simple and beautiful variety, aromatic fragrance ,and long florescence, simple maintenance and cold resistant.The Orchid traditional reproduction method mainly is minute reproduction. At the same time. Sedirea japonica's seed is extremely tiny, embryo is undifferentiation, and does not contain the endosperm, so the germination rate is very low. Along with tissue culture technology development, reproduces using the asepsis sowing seeds way, germination percentage will be improved in large scale. But the study on the technology of Sedirea japonica tissue culture has not been found yet in china. The Sedirea japonica's embryo were used as material to establish a rapid technique system of micropropagation, and researched its Tissue culture and fast reproduction technology. Studies emphatically its embrionic culture and differentiation of Protocorm like bodies ,and the best culture method. The main technology point is as follows:1,Embryo Culture:inorganic salt density and hormone in culture medium influence embryo's germination ,and joins 0.5mg/L KT in the 1/4MS culture medium effect to be best, the germination rate may reach above 70%; In the culture medium joins the coconut water, extracts of potato, the peptone and the activated charcoal is advantageous to embryo's germination; 2,The multiplication of the PLB: 1/3MS+6-BA2mg/L+NAA0.1mg/L+ coconutwater 10%+ peptone 2g/L+AC2g/L+ sucrose 20g/L+ agar-agar 7g/L.3,The differentiation of the PLB: 1/3MS+6-BA 10 mg/L+NAA0.5mg/L was the best medium. Each explant can obtain 2.8 shoots averagely.4,Seeding stage: the medium for the elongation of shoots and leaves was 1/2MS+IAA0.2mg/L+6-BA 6mg/L+ AC 1mg/L ;the medium for the rooting of shoots was 1/2MS+IAA0.5mg/L++6-BA 1mg/L+AC 2g/L.5,Active carbon was effective for preventing the explants from browning to a certain extent, 2mg/L was the best initiation and multiplication concentration, 6,For gardening of plantlets , the soil of the mess was the suitable material. The survival rate of plantlets could be up to 100%.
