| BACKGROUND:Atherosclerosis is the first cause of disability and death for Type 2 diabetic mellitus (T2DM).There is not any way to cure atherosclerosis. The development of preventative measures of atherosclerosis is currently a big concern for medical people.The formation and activation of atherosclerotic lesions is a multifaceted process involving many determinants,.among these factor,hyperglycemia product oxidative stress which induce the over express of adhesion molecules,such as:intercellular adhesion molecules (ICAM-1), E-selectin or imflamation's cytokinese, Such as:TNF-a IL-6 et al.Adhesion molecule and cytokines over express is the most important sign of endothilum lesion.Which is the most important start step for AS.so ,impress the oxidative LDL and inhibite the express of adhesion molecule and cytokines is beneficial for provention the development of AS early.Current evidence strongely suggested that soy isoflavone (special:Genistein,,G and daidzein,D) ascorbate(VC)and a-tocopherol (VE) protect against atherogenesis by inhibiting LDL oxidation. and by limiting the celluar responses to oxidized LDL,in particular adhesion molecule expression .But most soy isoflavone's conclution result from the high dose isoflavone. Lower dose of Gand D,the weaker of isoflaveone's antioxidant effects.It was thinked that give the isoflavene alone,the anti-oxygenation is very weak. Wohlquist MT et al study shown:The effect is more beneficial when intake other antioxidants company with isoflavone than intake other antioxidant alone For this,We try to use the plentifulness natural antioxidant VC and VE company with isoflavone to attain most prevention effect on DM complication AS.Object:First:Provide the basis for the antioxidant application clinically then prevent the AS development and progression.Second:Further demonstrated the mechanism of DM chronic complications .present that examinate early DM patient's adhesion molecule and cytokines is helpful for discovers and evaluate the atherosclerotic lesions.Finally:Further pull our country's soybean consumption confirmed the effects of soy isoflavone on prevent the development and progression of AS,can increase the new goods level of soybean food in domestic and international,and can certainly obtain the good economy and social performancing.Method:The present study was divided into two phases. In the first phases. We detect the LD50 of isoflavone to evaluate the toxincity strength.kind and the possibility's target organ to deter wether the isoflavones are safely be eatten by human.In the second phases,the oral glucose tolerance test (OGTT),with four grounp of T2DM patient (n=8~10)and a grounp of normal human. Among the four DM grounp .besides to OGTT three grounp was intervented with high does mixture (contain:isoflavone 300mg,VC 100mg.VE 800IU);or middle does mixture (contain:isoflavone 150mg.VC 500mg.VE 400IU);or low does mirture (coutain:isoflavone 50mg.VC 100mg.VE 50IU). examinating the coucentration of sICAM-1,E-selectin,TNF-a and IL-6 at fasting and postprandial 1h,2h,4h and6h. and evaluating these in detail.T2DM patient in -patient who confirrm to strict roll staundart and to contral the sex and the obeses interference through two layering draw samples .Result:During the OGTT, circulating sICAM-1,E-selectin,TNF-a plasma levels significantly increased in both diabetic control grounp (B grounp ) (P<0.01) and normal subjects (A grounp ) (P<0.05); Dm grounp with Low dose of mixture (C grounp )have the same exchange(P<0.05) .But the two DM grounp with middle dose and high dose mixture (D grounp and E grounp ) abolished this phenomenon. isoflavone company with VC,VE administered significantly decreased circulating ICAM-1 and E-selectin plasma levels in diabetic patients Except TNF-a,IL-6.But D grounp and E grounp have no significate.
