Expression Of P-selectin And MMP-9 On Vascular Epithelium In Smoked Rat Brain

The ischemia cerebrovascular disease ( ICVD) has been commonly seemed in the disorders of nervous system. The incidence and the mortality of the disease are high. There are lots of dangerous factors affecting the disease. Smoking has been confirmed independent factor in smoke. Studies have indicated that adhesion molecule and matrix metalloproteinase -9 had played an important role in the arteriosclerosis. They all join the appearance of the cerebral infarction.Materials and Methods1. Materials60 healthy Wistar rats supplied by Department of experiment animal of China Medical University were used. Monoclonal antibody of P -selectin, MMP -9 and SABC test box were from BOSTER Biotechnology CO. LTD. Cigarettes were purchased from markets.2. MethodThe Wistar rats were divided into 6 groups. (1) normal group ( n= 6) . (2) short - time and big - dose smoking group(n = 12). (3) long - time and big - dose smoking group (n = 12).(4) long - time and small - dose smoking group(n = 12).(5) giving - up smoking group (n =12). (6) control group ( n = 6 ) . We obtained brain following experiment ending and observed expression of smoked rat by im-munohistochemistry stain. The result was analyzed by spss 8. 0 software.ResultThe positive substance of P - selectin and MMP - 9 showed brown. P - selectin locates in the membrane of the vascular endotheli-al cell. MMP - 9 locates in the paste of the vascular endothelial cell. The expression of P - selectin and MMP - 9 in normal group and control group is weak. The significant different expression is observed in other groups. The positive cells of P - selectin are mainly on capillary posterior microvein and the positive cells of MMP - 9 are expressed on vascular epithelium cell and blood brain Barrier.DiscussionThe ischemic cerebrovascular disease (ICVD) is a disease that directly threatens people life. Epidemiology study has showed smoking is an important dangerous factor in ICVD. But the mechanism between smoking and cerebral infarction is still unclear. The study has demonstrated the increasing of the cerebral vessels intention and destroying of the vessel itself were the direct cause of the cerebral ischemia. The lesion of the endothelial cell was the first - emerged change responsi-ble for the following changes in this kind of cell of patients with ICVD. And it has been reported the cerebral infarction is a procession of inflammation. Adhesion molecules play an important role in this procession.Intercellular adhesion molecule is a kind of membrane glycopro-tein. P - selectin is closed associated with cerebral infarction. P - se-lectin is an adhesion molecule expressed on stimulated endothelial cells and palates. It is located in Weibel - Palade bodies of endothelial cells and a - granules or dense granules of platelets. In thrombosis it mediates the interaction of granulocytes with plates and with stimulated endothelium in the region of tissue injury and to promote granulo-cyte/endothelium fibrin deposition. It may be a receptor responsible for granulocyte " rolling" on endothelium.Nicotine of the cigarettes can injury the endothelial cell and stimulate endothelial to release adhesion molecule. It also stimulates contamination nerve to contract vessel. So that endothelial cell can express more P - selectin.Matrix metalloproteinase are a family of zinc - binding proteolytic enzymes which are capable of degrading components of the extracelluar matrix in a variety of physiological and pathophysiological conditions. Among MMPs, gelatinase B ( MMP - 9 ) is a major role to digest the endothelial basal lamina. It could be induced in a variety of cells in the central nervous system, including endothelial cells, microglia and neurons. It can be actived by plasmin. It' s collagen substance is IV type collagen formed based lamina. So MMP - 9 can help macrophage to go through endothelium to constitute arteriosclerosis and formed thrombosis further.
