Cerebral infarction (CI), because of its characteristics of high incidence,high morbidity, high recurrence rate and the high mortality, has become aserious diseases which is significantly harmful to human healthy. Traditionalrisk factors such as smoking, bad eating habits, high blood pressure,dyslipidemia, diabetes are associated with the occurrence of cerebral infarction.The genetic studies in recent years show that the occurrence of cerebralinfarction has genetic susceptibility. Atherosclerosis (AS) caused by cerebralinfarction is the most common type, also is an important pathological basis ofcerebral vascular disease, and endothelial cell injury is responsible forAtherosclerosis. E-selectin is a kind of cell adhesion molecules, It is highlyexpressed in the injury of endothelial cell surface. Thereby, we choose E-selectinA561C as candidate gene, through the study of the gene polymorphisms, toexplore the relationship between its and atherosclerosis cerebral infarction.Objective:To detect the E-selectin and A561C gene polymorphisms inpatients with cerebral infarction,and to study the relationship with cerebralinfarcton.Methods: We recruited199unrelated patients with cerebral infarction,admitted to the First Hospital of Jilin University, Changchun, China in theperiod between2011and2012for the case-control study. They were diagnosedas having cerebral infarction based on strict neurological examination, brainCT/MRI, which meet the International Classification of Diseases (NCHS2007).The124controls came from the physical examination center, the First Hospitalof Jilin University, in the period between2011and2012. To detect the E-selectin and A561C gene polymorphisms in the two groups by PCR-RFLP,SNPrs5361.Results: Three genotypes (AA, AC, CC) were found, There was significantdifferent in frequencies of allele (AA,AC,CC) and genotype(A,C) in E-selectinA561C polymorphism between CI group and control group (P<0.05); Relativerisk analysis indicated that the risk of cerebral infarction in C allele was5.4times higher than that in A allele(OR=5.4, χ2=17.887, P<0.05,95%CI:2.47-11.8). And the frequency of AC+CC genotypes in patients group hadsignificant higher than control group (P=0.00).Conclusions: E-selectin A561C polymorphism is associated withinfarction cerebral,and C allele may be the inherent predisposing gene forinfarction cerebral.
