Objective To detection the change of serum E-selectin and S128R gene polymorphisms in Han patients with infarction cerebral, and to study the relationship with infarction cerebral. Methods According to the condition of being selected, we selected 73 patients with the first ever acute cerebral infarction as cerebral infarction group and 85 patients without ischemic cerebrovascular disease were selected as normal control group. According to whether there are complications,we divided the 73 cerebral infarction patients into three subgroup:no complications group, hypertension group, combined hyperlipidemia group.Respectively, there were 15,35,23 patients in each group. In 24h after attacked the cerebral infarction, all the patients underwent skull CT(or MRI),and got fasting blood from Cubital vein; All the normal control group underwent the same examnations and got fasting blood from Cubital vein at physical examination. Genotypes of E-selectin were typed by polymerase-chain-reaction-restriction-fragment-length-polymorphism (PCR-RFLP).Enzyme-linked immunosorbent assay (ELISA) was used to measured serum E-selectin among each group. Results three genotypes(SS,SR,RR) were found. There was significant difference in frequencies of allele(SS,SR,RR) and genotype(S,R) in E-selectin S128R polymorphism between IS group and control group(P<0.05), relative risk analysis indicated that the risk of IS in R allele was 4.958(OR=4.985,95%CI:1.543-15.928) times higher than in S allele; Contrast to controls, the level of sE-selectin increased significantly (t=13.139,P<0.05), and IS patients with hyperlipidemia and hypertension had higher levels of serum E-selectin than those without the above risk factors(P<0.05).Conclusions E-selectin S128R polymorphism is associated with infarction cerebral, and R allele may be a risk factor for infarction cerebral dewelling in Lvliang district; the level of serum E-selectin increased in cerebral infarct, which correlated with hyperlipidemia and hypertension.
