Platelet plays a pivotal role in thrombosis, especially arterial thrombosis. Gene variants, taking effect on the function of platelet, might be a risk factor of artery disease, such as acute myocardial infarction, stroke. Recently some researches report that GPIIb-IIIa HPA-1 polymorphism is associated with ischemic stroke among European. And C807T polymorphism of GP I a/II a is a risk factor of stroke in Chinese. Owing to the different effect of genetic factors in different districts, it is indispensable to carry out detail researches among Chinese population, which will benefit for knowing the role of platelet membrane glycoprotein polymorphism in stroke as well directing the therapy of thrombosis disease especially stroke.The present researches focus on a series of relative study on GP I ba and GP III a polymorphism. Firstly, we apply a case-control mode and RFLP technique to detect kozak -5C/T polymorphism of 135 normal people and 95 stroke patients and use Logistic regression to analyze the relationship of gene and stroke. Result: 1. GP Iba gene kozak polymorphism -5C/C genotype is associated with stroke (OR= 4.49, 95%CI: 1.55-12.98, P<0.006). 2. The risk of-5C/C genotype on stroke is obviously increasing among groups lacking some common risk factors, especially in non-Hyperlipidemia group and female group. Consequence demonstrates that this genotype is an exclusive factor independent of other risk factors. 3. GP Iba gene kozak polymorphism and hyperlipidaemia have obvious combined effect on stroke.Kozak -5C/T, VNTR, HPA-2 polymorphism of GP Iba, which might be associated with arterial thrombosis, lie closely in the same chromosome. To eliminate the effect of VNTR, HPA-2 polymorphism on kozak polymorphism, we deploy the study on the linkage equilibrium of kozak -5C/T, VNTR and HPA-2 polymorphismResearch on the association of the platelet glycoprotein gene polymorphism and stroke Abstractand as well as the combined effect of the three polymorphism on stroke by using RFLP and SSCP technique. Result: 1. Neither GP Iba gene VNTR nor HPA-2 polymorphism is associated with stroke. 2.GP Iba gene kozak -5C/T VNTR, HPA-2 are not in linkage equilibrium. 3. D homozygous of VNTR polymorphism is in linkage with homozygous of HPA-2a. And kozak -5C homozygous is in linkage with homozygous of HPA-2a. Although aa/CC genotype is obviously different between control group and cerebral infarction group, there is no increasingly risk on onset of cerebral infarction compared with CC genotype of kozak -5C/T.Thirdly study on the effect of high prevalent GP IIIa Taq I polymorphism in Chinese individuals on stroke, aiming at finding another important genetic variants gene, we apply a case-control mode and RFLP and SSCP technique to detect GP IIIa Taq I polymorphism of 90 normal people and 90 stroke patient and use Logistic regression to analyze the relationship of gene and stroke. Result: 1. the GP IIIa Taq I polymorphism has no effect on stroke even in different layers. 2. SSCP technique is better than RFLP for its accurateness and low expense, which suggests that SSCP is up to be applied in investigation of polymorphism among large population.We fistly report GP Iba Kozak -5 C/T polymorphism is associated with stroke, which demonstrats that GP Iba Kozak -5 C/T polymophism is a new genetic variances gene of stroke among Chinese.
