Rhizoma pinelliae is usual traditional Chinese medicinal materials, but because of the difference in producing area and variety, the difference exists in the quality of rhizoma pinelliae. This work regards commonly used traditional Chinese medicinal materials rhizoma pinelliae as the research object, collects 16 samples, adopting UV absorption ,eletrophoresis, micellar eletronetic capillary chromatography (MECC) and RP-high performance liquid chromatography (HPLC) , we have carried on the differentiation research of the variety and quality of rhizoma pinelliae .This work provided convenience end scientific basis for the variety identification of rhizoma pinelliae in the real work.With UV absorption, the contents of the total protein in the fresh rhizoma pinelliae and other varieties had been determined . The basic principle is as follows: Tyrosine, tryptophan, phenylalanine and other residue in the protein molecule have the maximum absorption in 280nm wavelength. Because various kinds of proteins contain the tyrosine, so the absorbency in 280 run wavelength is an universal nature of protein . The absorbency and density of protein solution of certain density is in direct radio in 280nm, so it can be used to determined the content of total protein.. There is no reagent to join in the quantitative course, the protein is recoverable, and the method is simple and convenient.With isoelectrofocusing (IEF) electrophoresis and SDS polyacrylamide gel electrophoresis (SDS-PAGE) technology, the electrophoresis action of protein in fresh rhizoma pinelliae and other varieties have been studied. Isoelectrofocusing (IEF) technology is more advanced than PAGE .The samples of Chinese traditional medicine with protein composition can be analyzied and identified. This technology can form smooth pH gradient by adding ampholyte and make protein stop electrophoresis at the isoelectric points . In order to reduce expensive reagent consumption and finish IEF with the ordinary electrophoresis equipment, we have used improved IEF technology which is effectual and the reappearing is good. SDS (sodium dodecyl sulfate)- PAGE technology regards SDS as reducing agent which make protein samples reduce, there is a good linear relation between the reduced samples mobility and their molecular weight's logarithm(LogM) .Therefore, this technology can determine the molecular weight of the protein in samples .We have found : the IEF atlas of rhizoma pinelliae is stability and displays 8 characteristic bands,the isoelectric points (PI) of the bands have been determined; the SDS-PAGE atlases of rhizoma pinelliae and other varieties have obviousdifference ,the SDS-PAGE atlas of rhizoma pinelliae has 13 characteristic bands and the molecular weight of these characteristic bands have been determined by standard curve . So we can draw a conclusion that the method is reliable and steady, and the eiectrophoresis atlas of rhizoma pinelliae can be regarded as the differentiation basis of rhizoma pinelliae, rhizoma pinelliae PI value and the molecular weight of characteristic bands can offer the basis for isolation and purification of protein in rhizoma pinelliae. With micellar elefonetic capillary chromatography (MECC), the contents of benzoic acid in 14 samples of rhizoma pinelliae and other varieties have been determined. The capillary column is 62.5cm long and 75μm in internal diameter ;The operation voltage 25kv;test wavelength 200nm;column temperature 35℃;the mobil buffer is 30mmol.L-1 boric acid —15%methyl alcohol (pH9.5); the sampling is by pressure,the sampling time is 3 seconds. Results are as follows: the contents of the benzoic acid in pinelliae ternana from different areas is in 0. 022%-1. 568%; the contents of the benzoic acid in pinelliae pedatisecta from different areas is in 0. 087%- 0. 799%; the contents of the benzoic acl-1 in typhonium flageiliforme from different areas is in 0. 014%- 1. 409%.
