Objective. To study the quality and the suppressing effection to tumor of Pinellia plants.Methods. To determine the content of succinic acid by High Performance Liquid Chromatograpn (HPLC) and that of benzoic acid by High Performance Capillary Electrophoresis (HPCE), as well as to segregat protem by means of Isoelectric Focusing (IEF) and Sodium Dodecyl Sulfate Golyacrylamide Gel Electrophoresis (SDS-PAGE). To discriminate Pinellia DNA and its genetic polymorphism by Random Amplified Polymorphism DNA (RAPD) and estimate the content of protein. MTT reduction assay were applied to determine the suppressing effection to cultured Bel-7402 and HT-29 cells of Pinellia extracts and protein. Flow cytometry (FCM) were applied to estimate the promotion apoptosis of those extracts and protein.Results. The method of succinic acid content array by HPLC is assure that can segregate the peak of succinic acid and that the peak is symmetrical with straight and aclinic baseline. The method is resistant, repeatable and precise. However, the succinic acid content is diverse evidently from the origins, the gathering phases and the size of Pinellia. The method of benzoic acid content array of Pinellia by HPCE isestablished with small quality of samples required, efficient separation, acceptable resistant and repeatability. The content of Pinellia is different distinctly from one origin to another and the ratio of benzoic acid is 0.014% ~ 1.568%. The results of SDS-PAGE show that the same genus of different origin Pinellia reveals similar bands while the different genus exhibits different bands. The character chart array of Pinellia protein is established which can calculate the molecular weight of the bands. The protein of Pinellia can be separated and distinguished by IEF and that array shows good resistant and repeatability which can also distinguish some processed herbal such as ginger Pinellia. It is available that the proteins are separated and measured the isoelectric point (pi) of those protein bands. The RAPD array was applied to estimate the genie polymorphy of Pinellia .The result suggests that genie variety of Pinellia is not significant different from wildness to cultivation, while the number of genie polymorphy of wild Pinellia is two percent more than the fostered. The tiptop is wild Pinellia from Zhongxiang, Hubei, which indicate that origin is the most abundant of genie information and the best quality. The consequence of MTT and FCM to Bel-7402 and HT-29 intimates that the portion of organic acid and the extract of acetic ether exhibit ecumenic effect of suppressing tumor.
