ObjectiveThe centrosome,a small organelle located outside the nucleus of the cell, divides during cell division . From each centrosome microtubules grow into a spindle which is responsible for separating replicated chromosomes into the two daughter cells. Centrosome duplication is a key requirement for bipolar spindle formation and correct segregation of chromosomes during cell division. Without the correct number of centrosomes, human cells could not divide normally, resulting in a gain or loss of chromosomes within a genome which is known as aneu-ploid having intimate relation with unstable genome. In many kinds of tumor tissues and cancer cell lines can find abnormal centrosome and aneuploid, which is believed to be a critical cause of tumor formation through activating associated oncogenes and inhibiting some anti - oncogenes. It has been found some genes are responsible for centrosome replication. Their normal function ,structure and expression have important meaning for centrosome . STK15 just is one of these critical genes.STK15 (BTAK/aurora2) is a member of serine/threonine kinase gene family, which locates in 20q13. 2 . It encodes a kinase which has initiate relation with centrosome and it seems that STK15 is located in the centrosome. STK15 which is responsible for adjusting the replication of centrosome is little in the normal cell and mostly it plays the role by binding or cutting phosphate. However, it can be seen in a lot of abnormal of STK15 such as STK15 amplification, overexpression and so on in many sorts of tumor tissues and tumor cell lines. Much study suggest that STK15 is meaningful during most of cancers. It cancause three or more centrosomes appearencing,which will lead to the formation of aneuploid by STK15 amplification and overexpression.Laryngeal carcinoma is a cancer which has higher accidents in Northeast. The causes of this disease have not been elucidated. Our experiment is to analyze the role of STK15 in laryngeal carcinoma. Use differential PCR in carcinoma tissues and matched normal tissues of 40 patients to study STK15 amplification. Measure mRNA expression by RT - PCR with the same tissues. Test protein expression of STK15 by immunohistochemistry. In order to elucidate the role of STK15 clearly, analyzed the association among them three and the association of them all with kinds of clinical parameters of 40 patients such as age, sex. Our study will offer some scientific evidences for the causes of laryngeal carcinoma formation.Methods1. Use differential PCR in carcinoma tissues and matched normal tissues of 40 patients to study STK15 gene amplification.2. Measure mRNA expression by RT - PCR with the same tissues3. Test protein expression of STK15 by immunohistochemistry.Results1. In the 40 LSCC cases, STK15 amplification was detected in 14 patients (35% ); found 4 cases in 40 paried normal tissues ( 10% ). Analyzing data of the two teams by statistical test, STK15 amplification of the tumor team is obviously higher than that of normal control.2. STK15 mRNA overexpression was detected in 27 cases in the 40 LSCC patients(67.5% ) ; In the control team, tested 8 cases in 40 of all(20% ). It was clear that the STK\5 mRNA expression of LSCC is higher than paired normal tissues through statistication.3. In 29 cases out of 40 above LSCC cases, STK15 protein overexpressed.
