| Abstract: AIM To induce of PCD of leukemia cells is an important strategy in leukemia therapy.To find inducers of apoptosis which have lower side effect and obvious curative effect have been put to the front in new tumor therapeutic drug investigation and development. Better knowledge of cellular mechanisms that regulate apoptosis will be accompanied by further understanding of mechanism of apoptosis. We investigate the effect of glucosamine sulfate(GS) on chronic myloid leukemia K562 cells and the possible functions of Bcl-2 family proteins in the cathepsin D mediated apoptosis of K562 cells induced by GS. METHODS Electron photomicroscopy and Wright 's-Giemsa staining were used to study the morphologic evidences for apoptosis of K562 cells induced by glucosamine sulfate (GS). Cell cycle and Annexin V were detected by flow cytometry. The analysis of DNA Ladder was tested by agarose gel electrophoresis. Immunofluorescence was used to observe the translocation of cathepsinD and cytochromeC during the apoptosis. The expression of caspase-3, cathepsin D, bcl-2 bcl-xL, bid and bax in K562 cells treated by GS were detected by using western blot.
