cervical cancer is the second common malignant tumor inferior to galactophore cancer amog women throughout the world,its incidence is still at the first place in some developing country. With the development of common check and common therapy the incidence and mortality rate of cervical cancer obviously decline in some country or area,but the incidence rate of cervical cancer has a rising trend among young woman. Cervical intraepthelial neoplasia(CIN) is generic name of a group of cancer prophase lesion which has close relation with the tumor's invasion and reflects a continouse process in the occurrence of cervical cancer. Richart indicate the more higher degree of atypia is the more deeper of involved epithelium thickness,the more likely of invasive carcinoma ,contranly CIN of the lower grade reverse normal that is more likely. Prognostic factor of cervical cancer have many caurses,but among these factors clinical staging ,grade lymph node metastasis are important factors which affect prognosis.But until now we have no precise knowledge of the mechanism how cervical cancer happened .Like other human tumor the occurance and progress of cervical cancer are a series of genes change ,the inactivation of anti-oncogene play an important role in above-mentioned condition .In recent years, the abnormal transcription and expression of FHIT has close relation with the tumor's occurance and development in cervical cancer . Objective The purpose of this study is to aim directly at the close relation between cervical cancer and CIN and detect the expression of FHIT in different tissues by immunohistochemical method,which is to discover whether if there is significance in the expression of FHIT from normal tissue to malignant tissue.Meanwhile we analyze the relation between the expression of FHIT and pathological type, histological grade, clinical patholo-gic staging, lymph node metastasis and provide reference foundation to monitor lapse of CIN and prognosis of cervical cancer. Method To detect the expression of FHIT immunoh-istochemistry were used in cervical cancer tissue of 35 patients(squamous carcinoma 28,adenocarcinoma 7) and 39 EIN samples(gradeâ… 16,gradeâ…¡14,gradeâ…¢9) . All spacies come from the first hospital of jilin university (2003-2004) and were confirmed by pathology and which weren't drug treatment and radiation therapy beforesugery.Detecting the expression of FHIT in above-men-tioned cervical cancer and CIN by SP immunohistochemical method.Displacing the first antibody with PBS which become negative contrast. Making known positive expression tissue section of FHIT of mammary cancer as positive contrast. Observing results make use of double blind procedure . The expression product of FHIT show brown particles in cytoplasm or nucleus. Detecting and contrasting the positive expression of FHIT in normal self-tissue and tumor tissue. Refering to Greenspan'half quantitative method and scoring the distrbution,positive strength and positive rate of FHIT protein. First according to the degree of stain score zero, one ,two and three respec-tively .Second according to precent of positive cell score one ( or=51% ) respectively .When production of multipliction of precent of positive cell and degree of stain score 1~3 , FHIT express descend. while production score 4, FHIT express normal. Use software SPSS11.0 to analyze results of immunohistochemistry which is divided into groups according to different clinical pathological marker,and make use of X2 test to compare rate among these groups.Results 1. The FHIT expression of normal cervical tissue,CIN and cervical cancer tissue:The positive expression of FHIT protein submits to secondary strong pigmenting in normal cervical squamous epithel-ium,endocervix glandular epithelium and interstitial cell,lymphocyte,neutrophil,plasmacyte,et al in tu-mor.The expression of FHIT has a decreasing trend in normal cervical tissue,CIN and cervical cancer tissue . The reduced expression proportions of FHIT in cervical cancer is 57.14 %(20/35),20.51%(8/39) , there is significance among three groups(P <0.05). 2.The expression of FHIT in different cervical cancer tissue :(1). Reduced expression proportions of FHIT in squamous carcinoma is 67.86%(19/28) and 14.29%(1/7) in adenocarcinoma ,the former is obviously higher than the latter , there is significance between two groups(P <0.05);(2). Reduced expression proportions of FHIT in histological gradeâ… â…¡â…¢is 63.64%(7/11),53.33% (8/15), 55.56%(5/9) respectively,the differentce among grade â… â…¡â…¢is none of significance(P >0.05);(3). Reduced expression proportions of FHIT in clinical pathologic staging is 47.37% (9/19)in stage â… ,44.44% (4/9)in stageâ…¡a,75% (3/4)in stageâ…¡b ,there is no significance among three groups(P>0.05) ;(4). Reduced expressionproportions of FHIT with lymph node metastatic and no lymph node metastatic is 88.89% (8/9),46.15%(12/26)respectively, there is significance between two groups(P <0.05). 3. The expression of FHIT in different grade CIN tissue: Reduced expression proportions of FHIT in grade â… â…¡â…¢is 6.25%(1/16),28.57%(4/14),33.33%(3/9) respectively,the difference among grade â… â…¡â…¢is none of significance(P >0.05). 4. The expression of FHIT in squamous carcinoma and CIN tissue: Reduced expression proportions of FHIT in CIN is 20.51%(8/39) and 67.86%(19/28) respectively, there is apparent significance between two groups(P <0.01). Conclusion The expression of FHIT has a decreasing trend in normal cervical tissue,CIN and cervical cancer tissue .FHIT protein express decreased at a quantitive proportion in different grades of CIN ,and the proportion is higher following the increasing of lesion degree, there is no significance among three groups(P >0.05); The reduced expression of FHIT between CIN and squamous carcinoma ,between CIN and cervical cancer whose difference is of apparent significance(P <0.01); The reduced expression of FHIT in group squamous carcinoma is obviously higher than group adenocarcinoma, there is...
