The Expressions And Significances Of CyclinD1,p16,p21and Ki-67 In The Benign Prostatic Hyperplasia

Objective: To study the expressions and significances of CyclinD1,p16,p21 and Ki-67 in the benign prostatic hyperplasia (BPH) and explore the correlation during them. Methods: 80 specimens of benign prostatic hyperplasia patients were selected from Jiang Xi Provincial Hospital, surgical resected from 2000 to 2004, paraffin embeded tissues. Men 80 cases, age: 56~83 years old, average 68.3 years old. According to the different ratio of different histological type tissues proliferated ,we divieded them to 3 groups: 1. main of glandular proliferation 30 cases; 2. main of fibrous proliferation 30 cases; 3. main of smooth muscular proliferation 20 cases. Normal prostate (NP) tissues 20 cases as control group, autopsy specimens, paraffin embeded tissues, obtained from the department of anatomy of Jiang Xi Medical College. Men 20 cases, age: 22~43 years old, average 35.4 years old. CyclinD1,p16,p21 and Ki-67 were studied by using tissue chip technique and the S-P immunohistochemical methods. All datas were tested by X2 and categorical variable association. Results: In the 30 cases main of glandular proliferative BPH and 20 cases NP epithelia, the positive rates of CycinD1 were 16.7% and 10.0%, there were no significant differences between them, P>0.05; the positive rates of p16 were 36.7% and 10.0%, P<0.05; the positive rates of p21 were 23.3% and 55.0%, P<0.05; the positive rates of Ki-67 were 73.3% and 35.0%, P<0.01. In the 30 cases main of fibrous proliferative BPH and 20 cases NP fibres, the positive rates of CyclinD1 were 23.3% and 15.0%, there were no significant differences between them, P>0.05; the positive rates of p16 were 33.3% and 20.0%, there were no significant differences between them ,P>0.05; the positive rates of p21 were 43.3% and 55.0%, there were no significant differences between them, P>0.05; the positive rates of Ki-67 were 56.7% and 20.0%, P<0.05. In the 20 cases main of smooth muscular proliferative BPH and 20 cases NP smooth muscles, the positive rates of CyclinD1 were 10.0% and 5.0%, there were no significant differences between them, P>0.05; the positive rates of p16 were 30.0% and 15.0%, there were no significant differences between them ,P>0.05; the positive rates of p21 were 35.0% and 45.0%, there were no significant differences between them, P>0.05; the positive rates of Ki-67 were 55.0% and 15.0%, P<0.01. In the BPH epithelia,fibres and smooth muscles, the positive rates of CyclinD1 were 16.7%,23.3% and 10.0% respectively, there were no significant differences among them, P>0.05. the positive rates of p16 were 36.7%,33.3% and 30.0% respectively, there were no significant differences among them, P>0.05. the positive rates of p21 were 23.3%,43.3% and 35.0% respectively, there were no significant differences among them, P>0.05. the positive rates of Ki-67 were 73.3%,56.7% and 55.0% respectively, there were no significant differences among them, P>0.05. In all the 80 cases of BPH, the total positive rates of CyclinD1,p16,p21 and Ki-67 were 17.5%,33.8%,33.7% and 62.5% respectively, between CyclinD1 and Ki-67, P<0.01, r=0.54; between p16 and Ki-67, P<0.01, r=0.43; between p21 and Ki-67, P<0.01, r=0.36; between CyclinD1 and p16, P<0.01,r=0.28; between CyclinD1 and p21, P<0.05, r=0.23; but between p16 and p21, P>0.05. Conclusion: In the epithelia,fibres,and smooth muscles of BPH and NP, the expressions of CyclinD1 were rare, though it occasionally expressed in the different typical tissues of BPH, the expressions of CyclinD1 had no correlation with BPH. The expressions of p16 in the epithelia of BPH were higher than in NP, and the positive expressions of p16 most in the basilar cells. Theexpressions of p21 in the epithelia of BPH were lower than in NP. In the epithelia,fibres,and smooth muscles ,the expressions of Ki-67 in BPH were higher than in NP. The expressions of CyclinD1,p16,p21and Ki-67 in BPH had no correlation with histological types. In the 80 cases of BPH, the expressions of Ki-67,p16 ,p21 and CyclinD1 were successively reduced, the low expressions of CyclinD1 were closely related with high expressions of Ki-67; the high expressions of p16 were positively related with high expressions of Ki-67; among p21 and Ki-67,CyclinD1 and p16,CyclinD1 and p21, there were correlated with each other; but between p16 and p21, there was no correlation. In the checkpoint G1/S cell cycle regulators of BPH, the expressions of CyclinD1 had no correlation with BPH, the high expressions of p16 and low expressions of p21 were closely related with BPH, there was the most significantly epithlia proliferated in the BPH. The large-scale application of tissue chip is available to test the researched specimens efficiently, the method has quickness,convenience,economy and accuracy characteristics.
