Objective:To observe the expression and change of Treg cells,Th17 cells and their related cytokines Foxp3,RORyt,IL-10 and IL-17 in the tissues of benign prostatic hyperplasia(BPH),and to preliminary explore the expression and significace of Treg/Th17 cells imbalance and related cytokines in benign prostatic hyperplasia.Methods:40 Sprague-Dawley(SD)male rats(8-week-old)weighing 300g±20g were randomly divided into control group,BPH model group,retinoicacid intervention group(RA-intervention group)and cyclosporine A intervention group(CsA-intervention group).The BPH model of SD rats was established by subcutaneous injection of testosterone propionate,and treated with retinoic acid and cyclosporine A separately.The wet weight of prostate of SD rats was weighed,the volume of prostate was measured by drainage method,the organ coefficient of prostate was calculated.The pathological changes of prostate tissue of SD rats were observed,the ratio of Treg and Th17 cells to CD4 cells was detected seperately by flow cytometry.The protein expression of Foxp3 and ROR?t in prostate tissue were detected by Western blot.The expression of IL-10 and IL-17 in peripheral blood was detected by enzyme-linked immunosorbent assay(ELISA).Result:1.Pathological observation of prostate tissue in rats:in the control group,the tissue structure of prostate was normal,the arrangement of epithelial cells was regular,and the size of glandular cavity was the same.In the retinoic acid intervention group(RA-intervention group),the prostate tissue hyperplasia was not obvious,the number of glands was more in quantity,the distribution was uneven;some of the glandular cavities were narrowed,the lumen margin was regular,some of the epithelial cells were high columnar,with a few inflammatory cells infiltration.In the BPH model group,the prostate tissue was obviously hyperplastic and the structure was destroyed;it could be seen that the number of glands increased with dense arrangement,the glandular cavity shrank,the epithelial cells proliferated,showing high columnar;the layers increased with crowded arrangement,the papillary structure increased obviously,the stroma was hyperemia,with different degrees of inflammatory cell infiltration and local fibrous hyperplasia.Compared with the BPH model group,the destruction of prostate tissue in the cyclosporine A intervention group(CsA-intervention group)was more obvious,the number of glands increased more,the glandular cavity shrank more seriously,the epithelial cells showed obvious stratified high columnar changes,disordered arrangement;obvious papillary structure was formed with a large number of fibrous tissue hyperplasia and a large number of inflammatory cells infiltration.2.The expression of Treg cells in control group was higher than that in BPH model group(P=0.011)and CsA-intervention group(P=0.000),but there was no significant difference compared with RA-intervention group(P=0.998).The expression of Treg cells in BPH model group was lower than that in RA-intervention group(P=0.017),but higher than that in CsA-intervention group(P=0.031).The expression of Th17 cells in control group was lower than that in BPH model group(P=0.046)and CsA-intervention group(P=0.000),but there was no significant difference compared with RA-intervention group(P=0.928).The expression of Th17 cells in BPH model group was higher than that in RA-intervention group(P=0.012),but lower than that in CsA-intervention group(P=0.027).3.The expression of Foxp3 transcription factor in control group was higher than that in BPH model group(P=0.016)and CsA-intervention group(P=0.000),but there was no significant difference compared with RA-intervention group(P=0.992).The expression of Foxp3 transcription factor in BPH model group was lower than that in RA-intervention group(P=0.030),but higher than that in CsA-intervention group(P=0.045).The expression of RORyt transcription factor in control group was lower than that in BPH model group(P=0.049)and CsA-intervention group(P=0.000),but there was no significant difference compared with RA-intervention group(P=0.962).The expression of RORyt transcription factor in BPH model group was higher than that in RA-intervention group(P=0.017),but lower than that in CsA-intervention group(P=0.010).4.The expression of IL-10 in control group was higher than that in BPH model group(P=0.040)and CsA-intervention group(P=0.000),but there was no significant difference compared with RA-intervention group(P=0.983).The expression of IL-10 in BPH model group was lower than that in RA-intervention group(P=0.014),but higher than that in CsA-intervention group(P=0.036).The expression of IL-17 in control group was lower than that in BPH model group(P=0.012)and CsA-intervention group(P=0.000),but there was no significant difference compared with RA-intervention group(P=0.965).The expression of IL-17 in BPH model group was higher than that in RA-intervention group(P=0.042),but lower than that in CsA-intervention group(P=0.011).Conclusion:In the BPH model group,the proportion of Treg cells decreased and the proportion of Th17 cells increased,resulting in an imbalance in the proportion of Treg/Th17,accompanied by a decrease in the expression of Foxp3 and IL-10,and an increase in the expression of RORyt and IL-17.With the increase of the degree of prostatic hyperplasia in rats,the above changes were more obvious.The ratio imbalance of Treg/Th17 cells may play an important role in the pathogenesis of BPH.
