Objective The article investigated the function and mechanism of Qi Ju capsule on the effect of the left ventricular hypertrophy (LVH), and provided an experimental basis for further developing and using this prescription in clinic through observing the effect of Qi Ju capsule on the blood pressure (BP), left ventricular weight (LVW) to body weight (BW) ration (LVW/BW), the intracellular Ca2+ concentration ([Ca2+]i) of cardiomyocytes, and the expression of c-myc mRNA in cardiomyocytes tissue in spontaneous hypertension rats (SHR).Methods Thirty two SHR were randomly divided into four groups: Qi Ju capsule group "A group" (8 rats), benazepril group "B group" (8 rats), Qi Ju capsule and benazepril group "C group" (8 rats), model group "D group" (8 rats); and other eight SD rats were the normal contrast group "E group". The respective dose and drugs were given to the treated groups, and the same dose of distilled water was given to the normal and model groups by stomach rearing once daily for 12 weeks. During the experiment, blood pressure was measured every week and the general condition was observed. After therapy, blood pressure curve was drawn, left ventricular weight to body weight ration(LVW/BW), level of [Ca2+]i and the expression of c-myc mRNA in cardiomyocytes tissue of each group were tested and the microstructure of cardiomyocytes was observed. All the results were analyzed with SPSS 11.5.Results Qi Ju capsule could reduce blood pressure, the left ventricular weight, the level of [Ca2+]j, the expression of c-myc mRNA in cardiomyocytes tissue of SHR(P<0.05) and made the microstructure of cardiomyocytes stable. Compared with benazepril, the function of Qi Ju capsule in improving general condition was stronger, the function in lowering the left ventricular weight, the expression of c-myc mRNA and making the microstructure of cardiomyocytes stable was similar (/^>0.05), but the function of decreasing blood pressure and level of [Ca2+]i was weaker ( P<0.05 ) and the effect on making the microstructure stable was similar. In additional, the function of group C was the best in the three treated groups. Compared with benazepril, the effect of group C on reducing [Ca2+]; was better C/^0.05) , and its function of making the microstructure stable and improving general condition was stronger. Some trend still can be caught although there was no significant difference in the other tests.Conclusions (1) Qi Ju capsule could prevent hypertensive LVH. (2) The mechanism of Qi Ju capsule on preventing LVH may be related with lowering blood pressure, reducing [Ca2+]j concentration and down-regulating the expression of c-myc. (3)The effect of both Qi Ju capsule and benazepril on preventing LVH was more better than Qi Ju capsule or benazepril only.
