Expression And Significance Of P21~(WAF1/CIP1) And P27~(KIP1) In Epithelial Ovarian Carcinoma

Objective: To investigate the expression of P21 and P27 protein in epithelial ovarian carcinoma ,and the correlation between their expression and clinical pathological parameters of epithelial ovarian carcinoma.Methods: We compared the expression of P21 and P27 protein in 30 cases of epithelial ovarian carcinoma to 10 cases of normal ovarian tissue and 10 cases of benign ovarian tumor (control groups), immunohistochemistry technique was utilized to examine the expression of P21 and P27 protein in the above ovarian tissues. Nuclear staining of P21 and P27 proteins were positive cells. Three semiquantitative classes were used to describe the number of positively stained cells: (-) none or < 5% of tumor cells positive;(+)between 5 and 49% of tumor cells positive;(++)≥50% of tumor cells positive;(++) was strong positive. X~2 test was used for enumeration data and rank sum test was used for ranked data.Results: (1) The positive rates of P21 were increasingly decrease in normal ovarian group, benign ovarian tumor group and in epithelial ovarian carcinoma group,which were 100.00%, 90.00% and 63.33% respectively;strong positive rates were 40.00%, 20.00% and 16.67%. Positive and strong positive rates of epithelial ovarian carcinoma group were lower than those of other two groups, the difference was obviously (P<0.05). The positive rates of P27 in normal ovarian group, benign ovarian tumor group and in epithelial ovarian carcinoma group were 90.00%,80.00% and 36.67% respectively, strong positive rates were 30.00%, 20.00% and 6.67%. Positive and strong positive rates of epithelial ovarian carcinoma group were lower than those of other two groups, (p<0.01). (2) The expression of P21 protein and P27 protein in epithelial ovarian carcinoma was associated with clinical stage (P<0.05).(3) Positive correlation was found between P21 expression and P27 expression (P=0.038).Conclusion: (1) Expression of P21 and P27 protein in epithelial ovarian carcinoma were downregulated and oppositely related to the clinical stage. These results suggested that the abnormal expression of P21 protein and P27 protein may promotetumorigenesis.(2) A significant positive correlation was fond between P21 and P27 protein expression levels. It may suggest that P21WAF1/CIP1cooperate with P27HP1 in tumorigenesis.
