Objective: To investigate the expression of tumor suppressor gene PTEN and anti-apoptosis gene Survivin in gliomas as well as explore the correlation between them.Methods: The expression of PTEN and Survivin gene in 45 cases of brain glioma were detected with immunohistochemistry S-P method and correlation between them were analyzed. All the cases did not receive radiotherapy or chemotherapy.6 cases of normal brian tissue were used as control.Results: 1 .The positive rate of the expression of PTEN in gliomas and normal tissues were 53.3% (24/45) and 100% (6/6). There was statistical significance (P<0.05) between them.From gliomas of grade I-II to III-IV, PTEN positive expression decreased gradually,the positive rate were 67.9% (19/28) and 29.4% (5/17),they were statistical significance(P<0.05).2.The positive rate of the expression of Survivin in gliomas and normal tissues were 55.6%(25/45)and 0.0%(0/6).There was statistical significance(P<0.05) between them. From gliomas of grade I-II to III-IV,Survivin positive expression raised gradually, the positive rate were 35.7%(10/28) and 88.2 %(15/17),they were statistical significance(P<0.05).3.There were 7 cases Survivin-positive of PTEN- positive,whereas 18 cases PTEN-negative of Survivin-positive. The expression of PTEN in gliomas is inversely correlated with the expression of Survivin.Conclusions: 1. The abnormal expression of PTEN was closely associated with glioma's differentiation grade,and played an important role in the staging of gliomas. 2. The abnormal expression of Survivin was closely associated with glioma's differentiation grade,and played an important role in the staging of gliomas.3. Combined inspection of them can be used as indicators to determine the glioma's degree of malignancy and prognosis.4. There was an inversely correlation between the expressions of PTEN and Survivin in gliomas. which showed that the mutation of PTEN and second activation weren't two isolated events in the staging of gliomas. there might be some correlation between them.
