To study the distribution and sequence of gene expressions of Nestin at differentlocations and intervals after ischemic injury in rats, and to explore the activities ofnervous stem cells after ischemic injury which provides a theoretic base for treating thecerebral injury.Method30 adult male rats weights 270±30g are selected. Cerebral ischemic reperfusionmodel is made by intraluminal suture occlusion of middle cerebral artery. Rats aredivided randomly into operation group and control group. The operation group isdivided into several groups according to different intervals after ischemic reperfusion.Each group has six rats. So does the control group. SABC immunohistochemistrymethod is used for detecting the expression of nestin at different locations and intervalsof both operation group and control group.ResultsNestin has a broad expression in normal cerebral tissues. However, its expressionis weakly and few. Especially in subventricular zone, the nestin-positive cells has most.At 6h interval, the nestin-positive cells has had a few expression. At Id interval, thenestin-positive cells increase, especially in hippocampus dentate gyms. At 3d interval,the nestin-positive cells increase obviously, and the cells in penumbral region ofparietal cortex increased remarkably. At 7d interval, the quantity and morphologicalchanges of nestin-positive cells are mostly remarkable. That in hippocampus has most and the next is penumbral region, while in striatum it has fewer. It has significantdifference between each operation group and control group(P<0.01) . The expression ofnestin-positive cells at different locations is basically consistent after ischemic injury,while increasing of nestin-positive cells are various at different intervals. It has agradually decreasing trend in proliferation velocity unit hour in each location.ConclusionNestin has a broad expression in normal cerebral tissues. Its intension is low. Theexpression of nestin-positive cells at each location after injury increase consistently.The proliferation velocity of nestin-positive cells unit hour has a gradually decreasingtrend. It suggests that the fastest activated speed of nervous stem cell is in early state ofinjury.
